摘 要:目的:比较McCoy、Hela、Vero、HaCat四种细胞培养沙眼衣原体的易感性及药物敏感性试验结果。方法:将5 000和1 000 IFU的衣原体分别接种到McCoy、HeLa、Vero、HaCat细胞中,培养48和72 h,通过碘染色和单克隆荧光抗体染色法,观察衣原体包涵体的数量。比较四种细胞对衣原体的易感性和两种染色方法的敏感性。培养McCoy和HeLa细胞,进行10株衣原体临床株对5种抗生素的药物敏感性试验,观察衣原体的最小抑菌浓度(MIC)。结果:在衣原体感染后48 h和72 h,比较McCoy、HeLa、Vero与HaCat细胞对沙眼衣原体的易感性,差异均具有统计学意义(P<0.05)。在相同接种浓度下,碘染色观察McCoy和HeLa细胞中衣原体包涵体数量比HaCat细胞多,Vero细胞中包涵体数量最少;荧光染色时,McCoy和HeLa细胞中衣原体包涵体数量较多,HaCat细胞包涵体数量最少。衣原体感染48 h,碘染和荧光染色计数McCoy、HeLa和HaCat细胞内包涵体数量无明显差异(P>0.05),Vero细胞具有明显差异(P<0.05)。在McCoy和HeLa细胞中,5种抗生素对衣原体的最小抑菌浓度范围基本相同。结论:沙眼衣原体对McCoy细胞和Hela细胞有较高的易感性,碘染色观察Vero细胞中衣原体包涵体敏感性低,衣原体在McCoy和Hela细胞中的药敏试验结果一致。Objective:To compare four cells lines used for culture of Chlamydia trachomatis. Methods:McCoy, HeLa, Vero and HaCat cell lines were infected with sreovar D Chlamydia trachomatis with 5 000 and 1 000 informing inclusion unit (IFU) per well. After culture for 48 and 72 hours, the infected cells were used iodine and immunofluorescence stain to identify chlamydial inclusions. Then 10 clinical isolates of Chlamydia trachomatis were tested against azithromycin, erythromycin, doxycycline, ofloxacin and clarithromycin in McCoy and HeLa cells. Quantitative data for two detection methods were compared using t test. Multiple groups of quantitative data were compared using One-way analysis of variance. Results: When cells were infected with Chlamydia trachomatis for 48 and 72 hours, the number of inclusions in McCoy and HeLa cell were more than Vero and HaCat cells. The difference is considered to be significant(P 〈 0.05). For Vero cell, the inclusions were found more in immunofluorescenee stain than iodine stain( P 〈 0. 05). The range of minimal inhibitory concentration (MIC) for Chlamydia trachomatis against five antibioties was similar in McCoy and HeLa eells. The MIC90 was same for two cells. Conclusion:McCoy and HeLa are good cell lines for culture Chlamydia traehomatis and suitable for anti- biotic susceptibility testing in vitro.
