摘  要:WRKY蛋白是一类植物特异的转录因子家族,在植物响应病害及非生物胁迫中起重要作用。通过生物信息学方法,从玉米基因组中得到3个WRKY家族基因序列(ZmWRKY14-like、ZmWRKY25-like、ZmWRKY62-like),预测表明3个蛋白都定位于细胞核。荧光定量PCR分析表明,3个基因在玉米的不同器官中都有表达,但具有组织表达特异性。ZmWRKY14-like和ZmWRKY62-like在幼果中的表达量均显著高于在其他组织中的表达量,ZmWRKY25-like基因在叶、穗丝和幼果中的表达量高于在其他组织中的表达量。盐胁迫下,ZmWRKY25-like基因呈上调表达,ZmWRKY62-like基因呈下调表达;干旱胁迫下,ZmWRKY62-like基因出现下调表达;低温胁迫下,ZmWRKY25-like基因呈上调表达。结果表明,ZmWRKY25-like可能参与了植物对盐和低温胁迫的响应,ZmWRKY62-like基因则可能参与了植物对盐和干旱胁迫的响应。WRKY proteins are large family of plant-specific transcription factors ( TFs ), play essential roles in regulating plant responses to pathogens and abiotic stress. In this study, we identified three WRKY transcription factor ( TF ) genes (ZmWRKY14-1ike, ZmWRKY25-1ike, ZmWRKY62-1ike ) in maize by using bioinformatics method. Plant-mPLoe predicted that these three proteins were all localized in nucleus. Real-time qPCR assay showed that ZmWRKY14-like, ZmWRKY25-1ike, ZmWRKY62-1ike expressed in all tested maize tissues, but had differential expression. The expression levels of ZmWRKY14-like and ZmWRKY62-1ike in young fruit were significantly higher than those in other tissues, while the expression level of ZmWRKY25-like gene in leaf, spike and young fruit was higher than that in other tissues. The expression levels of three genes under different abiotic stress were also analyzed by real-time qPCR. The expression of ZmWRKY25-1ike was up-regulated, while ZmWRKY62-like was down-regulated under salt stress. The expression of ZmWRKY62-1ike was down-regulated under drought stress. Under low temperature stress, the expression level of ZmWRKY25-like was up-regulated. These results indicated that ZrnWRKY25-1ike may be involved into plant response to salt and low temperature stress, and ZmWRKY62-like may be involved into plant response to drought and salt stress.
