摘  要:目的探讨嗜酸性粒细胞趋化蛋白(eotaxin-3)基因+2497、+77位点单核苷酸多态性(SNP)与支气管哮喘(哮喘)易感性、类型的关系。方法选择哮喘患者196例(哮喘组)、经体检健康者196例(对照组),其中哮喘组嗜酸细胞性哮喘(EA)140例、非嗜酸细胞性哮喘(NEA)56例。采用聚合酶链反应-限制性片段长度多态性技术检测eotaxin-3 SNP,ELISA法检测血浆eotaxin-3水平。结果哮喘组eotaxin-3+2497位TG基因型频率、等位基因G的频率高于对照组(P均<0.05);两组+77位等位基因T的频率和各基因型频率比较差异无统计学意义(P均>0.05)。NEA患者+2497位TG型频率高于EA患者(P<0.05)。TG型哮喘患者血浆eotaxin-3水平低于TT型哮喘患者(P<0.05)。结论 eotaxin-3+2497突变(T>G)与哮喘易感性及类型相关,而eotaxin-3+77位点SNP与哮喘易感性及类型无关。Objective To investigate the relationships of eotaxin-3 + 2497 and + 77 single nucleotide polymorphism( SNP) with types and susceptibility of asthma. Methods We recruited 196 healthy control subjects( control group) and196 patients with asthma( asthma group),including 140 cease of eosinophilic asthma( EA) and 56 cases of non-eosinophilic asthma( NEA). The polymorphism,allele frequency and genotype frequency of eotaxin-3 + 77 C T, + 2497 T G were detected by PCR-RFLP. ELISA was used to detect the level of eotaxin-3 in plasma. Results The genotype frequency of eotaxin-3 + 2497 TG and allele frequency of eotaxin-3 + 2497 G in the asthma group were significantly higher than those of the control group( all P 0. 05). However,the allele frequency of eotaxin-3 + 77 T and genotype frequency of eotaxin-3 +77 showed no difference between the asthma and control groups( all P 0. 5). The frequency of eotaxin-3 + 249 7 TG genotype in NEA patients was significantly higher than that in EA patients( P 0. 05). The level of eotaxin-3 in TG genotype asthmatic patients was significantly lower than that in TT genotype( P 0. 05). Conclusions Eotaxin-3 + 249 7 mutation( T G) is associated,but eotaxin-3 + 77 SNP is not associated with asthma susceptibility,eosinophilic asthma and non-eosinophilic asthma.
