摘 要:目的:探讨Fas与Fas L在芪白平肺胶囊含药血清诱导低氧大鼠肺动脉平滑肌细胞(pulmonary arterysmooth muscle cells,PASMCs)凋亡中的作用。方法:芪白平肺胶囊含药血清和空白血清的制备。大鼠PASMCs的培养及a-SMA细胞免疫荧光鉴定。MTT法筛选最佳含药血清及最佳低氧时间。正式实验根据MTT结果分为常氧对照组,低氧模型组,最佳含药血清组,含药血清作用最佳低氧时间后,TUNEL-FITC/DAPI法检测各组PASMCs凋亡情况,Western Blot法检测各组PASMCs的Fas与Fas L蛋白表达变化。结果:与低氧模型组比较,芪白平肺胶囊含药血清可明显促进低氧大鼠PASMCs的凋亡,但细胞内Fas与Fas L蛋白表达较模型组无显著差异。结论:芪白平肺胶囊含药血清能有效抑制低氧大鼠PASMCs增殖作用,促进其凋亡,但Fas/Fas L死亡受体通路可能不参与芪白平肺胶囊含药血清诱导低氧大鼠PASMCs凋亡的调控。Objective : To investigate the role of Fas and FasL expressions in apoptosis of rat pulmonary artery smooth muscle ceils(PASMCs) under hypoxia induced by Qibai Pingfei Capsule Medicated Serum. Methods: Qibai Pingfei Capsule medicated serum and blank serum were prepared. Rat PASMCs were cultivated and identified by a -SMA immunofluorescence. The best Qibai Pingfei Capsule medicated serum and the best time of hypoxia were determined by the MTT method. According to the MTT experimental results, PASMCs were divided into normoxia control group, hypoxia model group and the best Qibai Pingfei Capsule medicated serum group. After the medicated serum affecting the best time of hypoxia,the TUNEL- FITC/DAPI method was used to check the apoptosis status of PASMCs and the Western Blot method was adopted to detect the protein expressions of Fas and FasL protein. Results: Compared with hypoxia model group, Qibai Pingfei Capsule medicated serum can effectively accelerate apoptosis of rat PASMCs under hypoxia, but there were no significant changes in the expression of fas/fasL protein. Conclusion : Qibai Pingfei Capsule medicated serum can effectively inhibit the proliferation and induce apoptosis of rat PASMCs under hypoxia. But the apoptosis of PASMCs may not occur independently of Fas/FasL death receptor pathway.
