摘  要:目的:研究加味黄芪补气汤对肿瘤患者外周血淋巴细胞的促增殖及活化作用。方法:以黄芪和苦参为主药,辅以适量人参、黄精及甘草,制备加味黄芪补气汤。分离5位肿瘤患者的外周血淋巴细胞,分别将补气汤、黄芪甲苷、氧化苦参碱作用于淋巴细胞。细胞计数法检测各组淋巴细胞增殖情况;ELISA法检测各组细胞培养上清中IL-2含量;ELISPOT法检测各组细胞分泌IFN-γ的情况。结果:加味黄芪补气汤浓度为2.5、5.0μg/m L(原药冻干物)时能够促进肿瘤患者外周血淋巴细胞增殖,其中浓度为2.5μg/m L时效果最好,与对照组相比具有统计学意义(P<0.01);黄芪甲苷浓度为2.0、4.0μg/m L时能够促进肿瘤患者外周血淋巴细胞的增殖,其中4.0μg/m L时效果最好(P<0.01);氧化苦参碱各浓度对肿瘤患者外周血淋巴细胞的增殖基本无影响(P>0.05);IL-2分泌水平检测显示,加味黄芪补气汤、黄芪甲苷、氧化苦参碱浓度分别为2.5、4.0、8.0μg/m L时,较组内其他浓度效果较好,与对照组相比均有统计学意义(P<0.05);IFN-γ的ELISPOT实验结果显示,加味黄芪补气汤、黄芪甲苷、氧化苦参碱浓度分别为2.5、4.0、8.0μg/m L时,阳性细胞计数最多,与对照组相比均有统计学意义(P<0.05)。结论:加味黄芪补气汤在一定浓度范围内能够促进肿瘤患者外周血淋巴细胞增殖与活化,其中黄芪的有效成分黄芪甲苷参与淋巴细胞的增殖与活化过程,而苦参的有效成分氧化苦参碱可能只参与活化淋巴细胞的作用。Objective : To study the effect of Modified Huangqi Buqi Decoction on proliferation and activation of tumor patients' peripheral blood lymphocyte. Method: The astragalus root and Sophora flavescens were the main drugs, supple- mented with the right amounts of ginseng, polygonatum and licorice to prepare Huangqi Buqi Decoction. Separate the peripheral blood lymphocytes of five tumor patients who were respectively given Huangqi Buqi Decoction, astragalus glycoside and oxidation of matrine. Cell counting method was used to detect the proliferation of lymphocytes. ELISA method was used to detect the IL - 2 content in the supernatant of cell culture. ELISPOT method was used to detect the secretion of IFN - γ in each group. Results : Huangqi Buqi Decoction can promote the proliferation of lymphocytes in peripheral blood of tumor patients when the concentration was at 2.5,5.0 μg/mL ( TC lyophilisates) and the best concentration was 2.5 μg/mL. Compared with the control group there was statistical significance (P 〈 0. 05 ). Astragaloside IV in the concentration of 2.0, 4.0 μg/mL can promote tumor patients with peripheral blood lymphocyte proliferation and the best coneentration was 4.0 μg/mL( P 〈 0. 01 ). The concentration of Matrine on tumor proliferation of peripheral blood lymphocytes had no effect ( P 〉 0. 05 ). Detection of IL - 2 secretion showed that concentrations of Huangqi Buqi Decoction, astragaloside and matrine at 2.5 μg/mL, 4 μg/mL and 8 μg/mL were better than the rest groups' and had statistical significance compared with those of the control group(P 〈 0. 05 ). ELISPOT results of IFN - y showed that concentrations of Huangqi Buqi Decoction, Astragaloside IV, matrine at 2.5 μg/mL, 4.0 μg/mL and 8.0 μg/mL had the most positive cell counts and were significant (P 〈 0. 05 ) compared with those of the control group.
