加味黄风汤对IgA肾病大鼠肾组织α-SMA及TGF-β表达的影响

摘　　要：目的:观察加味黄风汤对IgA肾病模型大鼠肾组织α平滑肌肌动蛋白(α-SMA)及转化生长因子β(TGF-β)的影响。方法:IgA肾病大鼠造模后,随机分为空白正常组、模型组、雷公藤对照组、加味黄风汤单倍剂量及双倍剂量治疗组,给药12周。检测大鼠尿液、生化、肾组织病理及α-SMA及TGF-β表达改变。结果:加味黄风汤治疗组尿蛋白水平低于模型组(P<0.05)。治疗组肾小球系膜区IgA沉积较模型组明显减弱,系膜基质及系膜细胞增生程度较模型组轻,免疫组化α-SMA及TGF-β表达强度较模型组弱(P<0.05)。加味黄风汤治疗组α-SMA及TGF-β蛋白表达明显下降(P<0.05),m RNA表达亦明显下降(P<0.05)。结论:加味黄风汤能减少尿蛋白,减少肾组织系膜区IgA沉积,减轻系膜细胞及系膜基质增生,下调α-SMA及TGF-β蛋白及m RNA的表达,保护肾脏功能。Objective：The aim of the experiment was to observe the effect of Modified Huangfeng Decoction on the expressions of α- SMA and TGF- β in the kidney of rats with IgA nephropathy（IgAN）. Methods：The rats were randomly divided into normal group （ N ） , IgAN model group （ M ）, control group of tripterygium （ T）, Huangfeng Decoction single dose group（S） and Huangfeng Decoction double dose group（D）. The rats were examined 24 h albuminuria,blood serum creatinine and pathological change of renal tissue. Results ： The level of urinary protein in the treatment group was lower than that in model group （P 〈 0. 05）. In the treatment group, the IgA deposition in the glomerular mesangial area was significantly weaker than that in the model group; the mesangial matrix and mesangial cell proliferation were less than those of the model group ; the expression intensity of α - SMA and TGF - β was weaker than that of the model group （ P 〈 0. 05）. In the treatment group, the expression of α - SMA and TGF - β protein was significantly decreased （ P 〈 0.05） and the expression of mRNA was significantly decreased （ P 〈 0.05 ）. Conclusion ： Modified Huangfeng Decoction can reduce urinary protein, the IgA deposition in the mesangial area, mesangial cell and mesangial matrix proliferation, lower the expression of α - SMA and TGF - β protein and mRNA and protect renal function.
