1,25-二羟维生素D_3对单侧输尿管梗阻大鼠肾间质纤维化调节性T细胞的作用研究

摘　　要：目的:探讨1,25-二羟维生素D3[1,25(OH)2D3]对单侧输尿管梗阻(UUO)大鼠肾间质纤维化调节性T细胞(Treg)的作用。方法:54只健康雄性wistar大鼠随机分为假手术组(A组)、UUO组(B组)、1,25(OH)2D3治疗组(C组)。每组于术后第1天、第3天、第7天分批处死6只,留取肾脏组织。用HE染色观察肾脏病理变化,采用免疫组织化学染色法测定肾脏人叉头框蛋白P3(FOXP3)、平滑肌肌动蛋白(α-SMA)的表达。结果:与假手术组相比,UUO组FOXP3表达水平显著降低,α-SMA表达水平显著升高,差异有统计学意义(P<0.05);与UUO组相比,1,25(OH)2D3治疗组FOXP3表达水平显著升高,α-SMA表达水平显著降低,差异有统计学意义(P<0.05)。结论:调节性T细胞可能参与了肾间质纤维化的发生及发展;1,25(OH)2D3作为免疫调节剂可通过上调调节性T细胞减缓肾脏纤维化的进展。Objective：To explore the effects of 1,25- dihydroxyvitamin D3 on regulatory T cells in unilateral ureteral obstruction（ UUO） rats with renal interstitial fibrosis. Methods：Fifty- four Wistar rats were assigned to healthy control group（ A group）,UUO group（ B group）,and 1,25- dihydroxyvitamin D3 treatment group（ C group）. After surgery for 1 day,3 days and7 days,6 rats were selected from every group randomly,and killed to get the renal. Renal pathological changes were observedby HE staining. The level of Forkhead Box ProteinP 3（ Foxp3） protein and alpha smooth muscle- actin（ a- SMA） were assayed by immunohistochemistry. Results：Compared with healthy control group,the level of Foxp3 protein was significantly lower,and the expression of a- SMA protein was significantly higher in UUO group（ P〈0. 05）. Compared with UUO group,the level of Foxp3 protein was significantly higher,and the expression of a- SMA protein was significantly lower in 1,25- dihydroxyvitamin D3 treatment group group（ P〈0. 05）. Conclusion：Treg cells may involve in the progress of the renal interstitial fibrosis. As an immunomodulator,1,25- dihydroxyvitamin D3 might inhibit the progress of renal interstitial fibrosis through increasing the level of Treg cells.
