摘 要:[目的]筛选适宜铁皮石斛繁殖的最佳方法。[方法]以铁皮石斛成熟蒴果和1年生植株茎段作为外植体,研究MS培养基中添加不同浓度激素对铁皮石斛种子和茎段诱导、增殖以及生根的影响,同时比较了传统玻璃兰花瓶与聚丙烯透明薄膜袋在铁皮石斛工厂化生产上的成本投入。[结果]适合铁皮石斛诱导的培养基为MS+1.0 mg/L 6-BA+120 g/L香蕉+35 g/L蔗糖+7.0 g/L卡拉胶;适合增殖的培养基为MS+1.0 mg/L 6-BA+0.05 mg/L NAA+120 g/L香蕉+35 g/L蔗糖+7.0 g/L卡拉胶;适合生根的培养基为1/2MS+0.80 mg/L NAA+120 g/L香蕉+20 g/L蔗糖+7.0 g/L卡拉胶。对于工厂化生产而言,选用蒴果作为外植体具有利于无菌体系建立、增殖倍数多和苗长势整齐一致等优点,聚丙烯透明薄膜袋可以代替传统的玻璃兰花瓶。[结论]该研究可为减少铁皮石斛组培苗的生产成本、缩短生长周期、获得大量优质组培苗提供一定的理论依据。L Objective ] To select the best method for propagation of Dendrobium officinale. [ Method] This experiment used mature capsule and annual stem segments of D. officinale as explants. The paper studied the effects of MS medium supplemented with different concentration hormones on induction, multiplication and rooting of seeds and stem segment. The paper also compared the cost of traditional vase and transparent polypropylene film bags on factory production. [ Result] The appropriate medium for the induction of D. officinale was MS + 1.0 mg/L 6-BA + 120 g/L banana + 35 g/L sucrose + 7.0 g/L carrageenan, the appropriate medium for the proliferation was MS + 1.0 mg/L 6-BA + 0.05 mg/L NAA + 120 g/L banana + 35 g/L sucrose + 7.0 g/L carrageenan, the appropriate medium for rooting was 1/2MS + 0.80 mg/LNAA + 120 g/L banana + 20 g/L sucrose + 7.0 g/L carrageenan. The paper also showed that using capsule as explants was in favor of establishing the sterile system, multiplication times and consistent seedlings. Transparent polypropylene film bags can replace traditional vase. [ Conclusion ] The study provides certain theoretical basis for reducing production cost of D. officinale tissue culture seedling, shortening the growth period and getting a lot of high-quality tissue culture seedling.
