中药复方WCA-P及其拆方对人胰腺癌BxPC-3细胞悬液裸小鼠皮下接种瘤的影响

摘　　要：目的:通过Bx PC-3细胞株悬液造成胰腺癌裸小鼠皮下接种瘤模型,研究中药复方WCA-P及其拆方对皮下瘤的影响。方法:以人胰腺癌Bx PC-3细胞株悬液对裸小鼠进行皮下接种成瘤,随机分为复方组、清热解毒组、健脾组、5-FU组和空白对照组,分别予以相应治疗药物,观察各组抑瘤率,并检测血清NK及CA199水平。结果:空白对照组小鼠非灌胃死亡率高,消瘦明显,中药干预小鼠非灌胃死亡率低,营养状况较好。实验结束时,空白对照组小鼠体质量明显轻于健脾组和复方组(P<0.05);清热解毒组小鼠体质量轻于健脾组、复方组和5-FU组,健脾组小鼠体质量重于复方组和5-FU组,复方组小鼠体质量重于5-FU组,但差异均无统计学意义(P>0.05)。空白对照组小鼠血清NK水平低于5-FU组、健脾组和复方组(P<0.01),复方组血清NK水平最高。各药物干预治疗组血清CA199水平均低于空白对照组(P<0.01)。结论:中药及腹腔化疗可以改善人胰腺癌Bx PC-3细胞悬液皮下接种瘤裸小鼠的营养状况和体质量;中药,尤其是含有健脾作用的中药复方可提高人胰腺癌Bx PC-3细胞悬液皮下接种瘤裸小鼠血清NK水平并降低CA199水平,并可能对人胰腺癌Bx PC-3细胞悬液皮下接种瘤裸小鼠的带瘤生存期有延长趋势。Objective： To the effects of Chinese herbal compound and its separated prescription on subcutaneous tumor by establishing nude mice model suffering subcutaneous injection tumor with BxPC-3 cell suspension of pancreatic cancer. Methods： Subcutaneous injection tumor of nude mice was inoculated by BxPC-3 cell suspension of pancreatic cancer, they were randomly divided into the compound group, heat-clearing and detoxifying group, spleen invigorating group, 5-FU group and blank control group, and given corresponding medicines, then tumor inhibiting rate, NK in serum and CA 199 level were observed in different groups. Results： Death rate of non-intragastric administration of mice in blank control group was high, emaciation was obvious, death rate of non-intragastric administration of mice intervened was low, the nutritional condition was good. By the end of experiment, body mass of mice in blank control group was lighter than that of spleen invigorating group and compound group （P〈0.05）; body mass in heat-cleating and detoxifying group lighter than that of spleen invigorating group, compound group and 5-FU group, body mass in spleen invigorating group more higher than that of whole recipe group, body mass in compound group more higher than that of 5-FU group, there were all no statistical differences （P〉0.05）. NK level in mice serum of blank control group was lower than that of 5-FU group, the spleen invigorating group, compound group（P〈0.01）, NK level of serum in compound group was the highest. Serum CA199 in different drug intervention groups were lower than that of blank control group significantly （P〈0.01）. Conclusion： TCM and intraperitoneal chemotherapy can improve nutritional condition and body mass of nude mice with subcutaneous inoculation tumor induced by BxPC-3 cell suspension of pancreatic cancer; Chinese herbs, especially compound of Chinese herbs for invigorating spleen can promote serum NK level of nude mice with subcuta- neous inoculation tumor induced by BxPC-3 cell susp
