摘 要:目的:通过测定和分析实验大鼠外周血干扰素γ(IFN-γ)和白细胞介素4(IL-4)的含量,观察自拟宣肺定喘汤对咳嗽变异性哮喘大鼠免疫功能的影响,探讨复方中药对咳嗽变异性哮喘机体的免疫调节机制,为中药开发提供实验依据。方法:采用卵蛋白和氢氧化铝激发致敏并用卵蛋白激发的方法复制CVA模型大鼠,随机分为6组,各组间予以相应治疗3周后腹主动脉取血,检测血清IFN-γ、IL-4水平。结果:自拟宣肺定喘汤可明显提高CVA模型大鼠血清IFN-γ的水平,降低血清IL-4的水平。结论:自拟宣肺定喘汤能借助抑制细胞因子IFN-γ、IL-4的水平,降低机体抗原和特异性抗体交联,抑制或阻断咳嗽变异性哮喘的早期发病过程,具有抗气道炎症及免疫调节作用,可能是中药复方治疗CVA的机制之一。Objective: To observe the influences of selfmade XuanFei DingChuan Tang on immune function of rats with cough variant asthma(CVA) and discuss immune regulation mechanism of compound Chinese medicine on CVA by measuring and analyzing levels of serum IFN-γ and IL-4 of peripheral blood in experimental rats, therefore, to provide experimental basis for TCM development. Methods: CVA rats model were duplicated by using OVA, sensitized and challenged aluminum hydroxide and challenged OA, they were randomly divided into six groups, blood of all groups were drawn from abdominal aorta after corresponding treatment of three weeks, levels of IFN-γ and IL-4 were detected. Results: Selfmade XuanFei DingChuan Tang can improve serum IFN-γ level of CVA rats model, lower the level of serum IL-4. Conclusion: Selfrnade XuanFei DingChuan Tang can lower cross-link of body antigen and specific antibody by inhibiting levels of IFN-γ and IL-4 of cytokine to inhibit and stop early progress of CVA, which could resist airway inflammation and regulate immunity, it may be one of mechanisms of compound Chinese herbs in treating CVA.
