摘 要:目的:研究采用MTT法检测不同浓度的熊果酸对大鼠胸主动脉外膜成纤维细胞增殖的影响。方法:解剖分离SD大鼠的胸主动脉,获取胸主动脉血管外膜。对血管外膜成纤维细胞进行培养、纯化、传代。采用TGF-β_1诱导血管成纤维细胞增殖,用不同浓度的熊果酸抑制细胞增殖,用MTT法检测不同浓度的熊果酸对TGF-β_1诱导血管外膜成纤维细胞增殖的影响。结果:随着熊果酸浓度的增加,UA组的OD值逐渐减小,血管外膜成纤维细胞的抑制率逐渐增加,且各组UA细胞抑制率比较,差异均有统计学意义(P<0.05),UA3组(UA浓度45μmol/L)的细胞抑制率作用最强。结论:熊果酸对TGF-β_1介导的胸主动脉外膜成纤维细胞增殖具有明显的抑制作用,该抑制作用随着熊果酸的浓度增加而增强。Objective: To study the effects of different concentrations of ursolic acid on the proliferation of rat thoracic aorta adventitial fibroblasts by MTT assay.Method: The thoracic aorta of SD rats were dissected, and the adventitia of thoracic aorta was isolated.The adventitial fibroblasts were cultured, purified and passaged. TGF- β1 was used to induce the proliferation of vascular fibroblasts, different concentrations of ursolic acid were used to inhibit cell proliferation, MTT method was used to detect the effect of ursolic acid on the proliferation of vascular adventitial fibroblasts induced by TGF- β1.Result: With the increase of ursolic acid concentration, the OD of UA group decreased gradually, the inhibitory rate of adventitial fibroblasts was gradually increased, and the inhibitory rate of UA cells in each group were statistically significant ( P〈0.05 ), UA3 group ( UA concentration 45 μL mol/L ) had the strongest inhibitory effect.Conclusion: Ursolic acid significantly inhibit TGF-β1 mediated proliferation of adventitial fibroblasts of thoracic aorta, the inhibitory effect is enhanced with the increase of ursolic acid concentration.
