HPLC法测定四味宁心胶囊中葛根素的含量及药效学研究

摘　　要：目的:测定四味宁心胶囊中葛根素的含量,并对其进行药效学观察。方法:采用HPLC法对处方中的葛根素进行含量测定。观察四味宁心胶囊对抗氯仿致小鼠心率失常及其对兔脑循环和脑代谢的影响。结果:四味宁心胶囊在0.5~2.0μg范围内,呈良好的线性关系。葛根素平均回收率为99.29%(RSD%=0.56%);四味宁心胶囊能明显改善氯仿致小鼠心率失常,降低小鼠室颤率及明显增加兔脑血流量,并显著提高兔脑耗氧量和葡萄糖摄取量,且呈剂量依赖性。结论:本法操作简便,重复性好,结果准确,快速,可作为四味宁心胶囊的质量控制方法;且具有抗心肌缺血的作用和具有改善脑循环和脑代谢的双重作用。Objective： To determine the content of puerarin in Siwei Ningxin capsule and study the phar- macodynamics. Methods： The content of puerarin in the Siwei Ningxin capsule was determined by HPLC; We ob- served the effect of Siwei Ningxin capsule on chloroform induced arrhythmia in mice and cerebral circulation and cerebral metabolism in rabbit. Results： The linearcorrelation of puerarin was good in the range of 0.5-2.0 μg and therecovery was 99.29% （RSD=0.56%）. Siwei Ningxin capsule improved the chloroform induced arrhythmia, reduced the rate of ventricular fibrillation in mice and significantly increased cerebral blood flow and elevated cerebral metabolic rate for oxygen and glucose in a concentra tion-response fashion. Conclusion： This method is simple, reproducible, accurate and rapid. It can be used as the quality control method of Siwei Ningxin capsule. Siwei Ningxin capsule shows function of against myocardial ischemia, improving cerebrocir culation and cerebr ome tabolism.
