摘 要:目的 探索碱性成纤维细胞生长因子 (b FGF)对庆大霉素 (GM)和缺血性肾损伤的保护作用。方法 实验分两部分 :GM肾毒性拮抗作用研究及缺血性肾损伤拮抗作用研究。结果 两部分研究中对照组或模型组均可见肾小管上皮细胞明显肿胀、变性和坏死 ;实验组的肾小管上皮细胞损伤较轻或形态基本正常 ,而且b FGF可明显降低血清 BUN、Scr、MDA含量和组织 MDA、NO含量 ,增强 Ca2 + -Mg2 + -ATP酶活性。结论 b FGF对庆大霉素和缺血性肾小管损伤有很好的保护作用。AIM\ To explore protective effects of basic fibroblast growth factor (bFGF) on gentamicin (GM) and renal ischemic injury. METHODS\ The experimental group was divided into 2 parts: Part Ⅰ, to research bFGF′s antagonistic effect on GM renal toxicity: experimental animals were guinea pigs, which were randomly divided into control and experimental groups ( n =10). For control group, GM was injected for 7 d with 80 mg/kg·d\+\{-1\} and injected N.S. for 6 d after GM treatment. For the experimental group, bFGF was added for 17 d with 2000 U/kg·d\+\{-1\} while treated with GM, also to keep injecting N.S. for 6 d after medicines treatment. Then sample renal tissues with optical and electric microscopes were observed. Part Ⅱ, to research bFGF′s antagonistic effect on renal ischemic injury: experimental animals were 40 Wistar rats, which were equally and randomly devided into Pseudo operation, Model, High bFGF and low bFGF groups. Before and after the operations, MAD, BUN, and Scr in rats′ blood were measured . For Model group, N.S. 10 min before ischemia was renal vein injected and then clamped off the bilateral renal arteries for 60 min, and took off clamps and gave N.S. For high and low bFGF groups, to replace N.S. in model group by 4000 U/kg and 2000 U/kg bFGF respectively; For pseudo operation group, the bilateral renal arteries didn′t be clamped off and treated without NS. Renal tissue and made pathological slices and renal tissue homogenate in 24 h, then Ca 2+ , Mg 2+ and ATP enzyme′s activity, NO, MDA, and protein content were observed. RESULTS\ In control or model group of two parts′ research, epithelial cells of renal tubules swelled, denatured and putresced; the same parts in experimental groups showed lighter or normal states. bFGF could reduce serum′s MAD, BUN, Scr, NO and MDA content; it could also improve the activity of Ca 2+ , Mg 2+ and ATP enzyme. CONCLUSION\ bFGF can well protect animals from GM toxicity and renal ischemic injury. bFGF′s antagonistic effec
