摘　　要：目的探讨CYP17基因多态性与子宫内膜异位症的相关性。方法选择2014年1月至2015年12月深圳市人民医院收治的子宫内膜异位症患者30例(子宫内膜异位症组),同时选取体检健康者30例作为对照组,采集2组受试者外周静脉血标本并提取DNA,设计引物,通过聚合酶链式反应扩增包括基因多态位点的片段,用Msp A1Ⅰ限制性内切酶进行酶切,酶切产物在琼脂糖凝胶上电泳,确定CYP17基因的基因型,分析CYP17基因多态性与子宫内膜异位症的相关性。结果子宫内膜异位症组患者的CYP17基因型TT、TC、CC频率分别为36.7%、46.7%、16.7%,对照组CYP17基因型TT、TC、CC频率分别为20.0%、43.3%、36.7%,子宫内膜异位症组患者CYP17基因的TT基因型频率显著高于对照组(P<0.05),CC基因型频率显著低于对照组(P<0.05);2组TC基因型频率比较差异无统计学意义(P>0.05)。子宫内膜异位症组患者CYP17等位基因T、C频率分别为60.0%(36/60)、40.0%(24/60),对照组受试者CYP17等位基因T、C频率分别为41.7%(25/60)、58.3%(35/60),2组受试者CYP17等位基因频率比较差异有统计学意义(P<0.05)。CYP17基因的TC基因型与子宫内膜异位症的患病风险无关联(P>0.05);CYP17基因携带TT基因型个体罹患子宫内膜异位症的风险增加(P<0.05)。结论 CYP17基因多态性可能与子宫内膜异位症有相关性,可能为该疾病的遗传易感基因,CYP17基因携带TT基因型个体罹患子宫内膜异位症的风险增加。Objective To investigate the relationship between CYP17 gene polymorphism and endometriosis. Methods Thirty patients with endometriosis were selected in Shenzhen People＇s Hospital from January 2014 to December 2015 as endometriosis group. Meanwhile,30 healthy subjects were selected as control group. The peripheral blood samples of the subjects in the two groups were collected and DNA was extracted. The primers were designed,and the gene fragments were amplified by polymerase chain reaction（ PCR）. The PCR products were digested with Msp A1Ⅰ restriction enzyme,and enzymatic cleavages were performed with agarose gel. The relationship between CYP17 gene polymorphism and endometriosis was analyzed. Results The TT,TC and CC genotypic frequency of CYP17 gene in endometriosis group was 36. 7%,46. 7% and 16. 7% respectively; and the TT,TC and CC genotypic frequency of CYP17 gene in control group was 20. 0%,43. 3% and 36. 7% respectively;the TT genotypic frequency of CYP17 gene in endometriosis group was significantly higher than that in control group（ P 0. 05）,and the CC genotypic frequency of CYP17 gene in endometriosis group was significantly lower than that in control group（ P 0. 05）,but there was no significant difference in the TC genotypic frequency of CYP17 gene between the two groups（ P 0. 05）. The T and C allele of CYP17 gene in endometriosis group was 60. 0%（ 36 /60） and 40. 0%（ 24 /60） respectively,and the T and C allele of CYP17 gene in control group was 41. 7%（ 25 /60） and 58. 3%（ 35 /60） respectively; there was significant difference in the allele frequency of CYP17 gene between the two groups（ P 0. 05）. The TC genotype of CYP17 gene was not associated with the risk of endometriosis（ P 0. 05）,the risk of endometriosis in the individual with TT genotype of CYP17 gene increased（ P 0. 05）. Conclusions CYP17 gene polymorphism may be associated with endometriosis,CYP17 may be a genetic susceptibility gene for endometriosis. The risk of endometriosis is increased in
