摘 要:目的:探讨槲皮素能否减轻紫外线照射诱导的人精子氧化损伤。方法:通过收集10例健康志愿者精液,密度梯度法离心分离精子,利用紫外线照射90 s诱导精子氧化损伤。实验组添加50μmol/L槲皮素培养,采用计算机辅助精液分析系统(CASA)检测精子活动力,吖啶橙(AO)试验检测DNA损伤,流式细胞术检测活性氧(ROS)以及TUNEL法检测精子凋亡。结果:CASA显示各组精子活动力没有明显改变,AO试验表明实验组精子DNA损伤减轻,流式细胞术结果显示实验组精子内ROS含量下降,同时凋亡也相应减少。结论:槲皮素通过降低ROS,减轻紫外线照射诱导的人精子氧化损伤,从而起到保护作用。Objective: To evaluate whether quercetin could reduce oxidative damage in human sperm induced by ultraviolet radiatiown b irradiation. Methods: Ten semen samples from healthy volunteers were collected, centrifuged and isolated by density gradient centrifugation method, and induced oxidative damage by using 90 seconds of ultraviolet radiation. Sperms were cultured 1 h in vitro after quercetin (50 μmol/L) was added, then collected to detect sperm viability and motility by computer-assisted semen analysis system(CASA). In the meantime, DNA damage of sperms was detected by acridine orange test, reactive oxygen species (ROS) content and sperm apoptosis were detected by flow cytometry. Results: UVB irradiation led to an increase in sperm DNA damage, ROS production and apoptosis, and quercetin could reverse it, but had no effect on sperm motility. Conclusion: Quercetin plays a protective role in human sperm oxidative damage induced by UVB irradiation mainly by reducing the ROS content.
