摘  要:目的研究玄参对内毒素(lipopolysaccharide,LPS)诱导的急性肺损伤(acute lung injury,ALI)大鼠的保护作用,并探讨其可能的作用机制。方法 60只SD雄性大鼠随机分为正常对照组、模型组、阳性药组、玄参高剂量组、中剂量组、低剂量组,每组10只。采用气管内滴注LPS的方法建立ALI动物模型,造模后12 h取材,观察大鼠肺泡灌洗液中白细胞数目、总蛋白浓度、肺湿干比及肺组织病理切片。结果玄参、阳性药能有效的降低肺泡灌洗液中白细胞数目、总蛋白浓度及肺的湿/干比(P<0.01);显著减轻肺泡结构破坏、肺水肿和炎症细胞浸润等病变。结论玄参能有效改善ALI的炎症反应,降低肺部受损情况,提示玄参对ALI有一定的防治作用。Objective To investigate the effects of Scrophularia ningpoensis Hemsl on lipopolysaccharide-induced acute lung injury(ALI) in rats,and explore the underlying mechanism against ALL Methods Adult male Sprague-Dawley rats were randomly divided into six groups with 10 rats in each group:(1) control group(saline),(2) LPS group,(3) LPS+dexamethasone(LPS+Dex,2 mg/kg),(4) LPS+SNH(9 g/kg),(5) LPS+SNH(4.5g/kg),(6) LPS+SNH(1.5 g/kg).The cell counting and protein concentration in the bronchoalveolar lavage fluid(BALF) was measured.The animal lung edema degree was evaluated by wet/dry weight(W/D) ratio.Pathological changes of lung tissues were observed by hematoxylin and eosin(HE) staining.Results The data showed that treatment with the WESNH inhibited LPS-induced inflammation:SNH reduced the total protein concentration and the number of polymorphonuclear leukocytes in BALF;histological studies demonstrated that SNH substantially inhibited LPS-induced neutrophils in lung tissue.Conclusion The results indicated that the SNH had a protective effect on LPS-induced ALI in rats.
