摘 要:目的优选蛭丹化瘀组方的水提醇沉工艺。方法以干膏得率为参考,以芍药苷、黄芪甲苷含量为指标,采用L_9(3~4)正交试验优选出最佳的水提工艺,采用单因素试验优选出最佳的醇沉工艺。结果优选出的工艺为8倍量水煎煮3次,每次1 h,提取液滤过,合并滤液,浓缩至相对密度为1.10~1.15(60℃)的药液,冷却至室温,加入适量乙醇使醇浓度达60%。醇沉后,干膏得率减少约5%。结论优选出的水提醇沉工艺条件稳定性好,合理可行,可为大生产提供参考。Objective The technique of water extraction and alcohol precipitation of Zhidanhuayu Formula was optimized.Methods With dry extract yield as the reference,Paeoniflorin and Astragaloside IV extraction contents as indexes,L9(3^4) orthogonal test was adopted to optimize the best water extraction process,the single factor test was adopted to optimize the best alcohol precipitation process.Results Optimal process was:extracted 3 times with 8 folds of the amount of water,1 hour for each time.The extracts were filtered,combined and concentrated to the relative density of 1.10~1.15(60℃),cooled to room temperature.Add suitable amount of alcohol to attain to the alcohol concentration of 60%.After alcohol precipitation,dry extract yield decreased about5%.Conclusion The optimized condition of water extraction and alcohol precipitation was of good stability,reasonable and feasible,which can provide a reference for the mass production.
