摘  要:[目的]观察比较不同频率电针治疗对完全弗氏佐剂(complete Freund’s adjuvant,CFA)致炎性痛大鼠患侧热缩足潜伏期(thermal paw withdrawal latencies,PWLs)和机械缩足阈(paw withdrawal thresholds,PWTs)的作用及其对血清中肿瘤坏死因子α(tumor necrosis factorα,TNF-α)、白细胞介素1β(interleukin 1β,IL-1β)和白细胞介素6(interleukin 6,IL-6)的调控,讨论电针对CFA致炎性痛大鼠PWLs和PWTs优效频率及作用机制的异同性。[方法]所有实验大鼠完全随机分为空白组(normal)、模型组(model)、2Hz电针组(2Hz EA)、100Hz电针组(100Hz EA)、2/100Hz电针组(2/100Hz EA)和假电针组(sham EA)。除空白组外,其余各组均建立CFA诱导的炎性痛模型。电针组选用双侧"足三里"和"昆仑"穴,0.5m A-1.5m A,30min/次。分别检测各组大鼠造模前、治疗前、造模后1d、3d、7d、14d治疗后患侧PWLs和PWTs。检测大鼠血清中TNF-α、IL-1β和IL-6的含量。[结果]模型组大鼠在造模后各时间点患侧PWLs和PWTs均低于空白组,且差异具有统计学意义(P<0.01)。连续治疗14d后,各电针组大鼠患侧PWLs均明显高于模型组和假电针组,且差异具有统计学意义(P<0.01),100Hz和2/100Hz电针组大鼠患侧PWTs明显高于模型组,且差异具有统计学意义(P<0.01,P<0.05),其中2/100Hz电针组大鼠患侧PWLs上升最显著,100Hz电针组大鼠患侧PWTs上升最显著。假电针治疗组大鼠患侧PWLs和PWTs在治疗后各时间点与模型组比无明显变化。模型组大鼠血清中TNF-α、IL-1β、IL-6的含量均高于空白组,差异具有统计学意义(P<0.01)。各电针组大鼠血清中TNF-α的含量均低于模型组,差异具有统计学意义(P<0.01);2Hz电针组和2/100Hz电针组大鼠血清中IL-1β的含量均低于模型组,差异具有统计学意义(P<0.05,P<0.01);100Hz电针组大鼠血清中IL-6的含量高于模型组和假电针组,差异具有统计学意义(P<0.01)。假电针治疗组大鼠血清中TNF-α、IL-1β、IL-6的含量与�[Objective] Observing the effects of different frequency electroacupuncture(EA)’s regulation on thermal paw withdrawal latencies(PWLs), mechanical paw withdrawal thresholds(PWTs) and serum proinflammatory cytokines α(TNF-α, IL-1β, IL-6) on rats with inflammatory pain induced by completing Freund's adjuvant(CFA), to discuss the difference of the effect and mechanism of EA between the PWLs and PWTs. [Methods] All rats were randomly divided into normal group, model group, 2Hz EA group, 100Hz EA group, 2/100Hz EA group and sham EA group. Rats were injected with CFA to establish the inflammatory pain model, except normal group. EA group(0.5mA-1.5mA) was applied to bilateral ‘ Zusanli’ (ST36) and‘ Kunlun’ (BL60) for 30min. The PWLs and PWTs were measured before model and before EA administration, at 1d, 3d, 7d, 14d after EA administration. The expression of tumor necrosis factorα(TNF-α), interleukin 1β(IL-1β) and interleukin 6(IL-6) in serum of rats were detected after 14 days’ treatment. [Results] PWLs and PWTs of rats in model group were significantly decreased, which were lower than that of the normal group( P〈0.01) at each time point. After 14 days’ consecutive treatment, PWLs of rats in EA group were significantly higher than that of model group and sham EA group( P〈0.01). PWTs of 100Hz and 2/100Hz EA group were significantly higher than that of the model group(P〈0.01, P〈0.05). PWLs of rats in 2/100Hz EA group were highest among that of all groups, while PWTs of rats in 100Hz EA group were highest among that of all groups. There were no significant changes of PWLs and PWTs between model group and sham EA group in each time point. The expressions of TNF-α, IL-1βand IL-6 in the serum of model group were significantly higher than those in the normal group (P〈0.01).
