摘 要:以小黑杨花药为外植体,进行花粉植株(即单倍体)的诱导及体系的优化。通过染色体压片及PCR方法进行检测,证明已获得了单倍体植株。同时,以pROKII为载体利用根癌农杆菌介导法对单倍体植株进行了遗传转化,分子检测结果表明,外源基因已成功整合入单倍体小黑杨中。本研究可为林木单倍体的诱导及遗传转化研究提供参考,也可为杨树及林木的育种方式提供新思路。In this study, the anther of Populus simonii x P. nigra was used to induce pollen plant (haploid plant), and the improved method for haploid plant induction was performed. The squashing method for chromosome preparation and PCR method were used, and the results confirmed that the haploid plant was obtained. The haploid plant was transformed with pROKII using Agrobacterium tumefaciens-mediated method, and the result showed the exogenous gene was successfully introduced into genome of haploid plant. The present study will contribute to research of haploid induction and transformation,and breeding of poplar and other trees.
