七叶一枝花对H_(22)荷瘤小鼠实体瘤生长及生存时间的影响

摘　　要：目的观察七叶一枝花对H_(22)荷瘤小鼠实体瘤生长及生存时间的影响。方法于小鼠右前肢腋窝皮下(腹腔)接种0.2 m L瘤细胞液建立H_(22)荷瘤小鼠实体瘤模型,造模后按体质量将小鼠随机分为模型组(灌胃蒸馏水20 m L/kg,每日1次),阳性组(腹腔注射环磷酰胺40 mg/kg,隔日给药1次),七叶一枝花高(2.4 g/kg)、中(1.2 g/kg)、低(0.6 g/kg)剂量组(每日灌胃给药1次),每组30只。接种H_(22)瘤细胞后次日开始给药,连续给药14 d。末次给药后24 h,称取小鼠体质量后将其处死,剖取瘤块称重,计算抑瘤率。于小鼠腹腔接种0.2 m L瘤细胞液建立H_(22)荷瘤小鼠腹水瘤模型,分组及给药同上,给药结束后继续观察,并记录各组小鼠生存时间,计算生命延长率。结果与模型组比较,阳性组及七叶一枝花高、中、低剂量组瘤重均显著降低,差异均有高度统计意义(P<0.01);七叶一枝花高剂量组瘤重明显低于中、低剂量组,差异均有统计意义(P<0.05或P<0.01)。与模型组比较,阳性组及七叶一枝花高、中、低剂量组生存时间均显著延长,差异均有高度统计意义(P<0.01);七叶一枝花高剂量组小鼠的生存时间明显长于低剂量组(P<0.05)。结论七叶一枝花可明显抑制H_(22)荷瘤小鼠实体瘤的生长,并延长H_(22)荷瘤小鼠的生存时间。Objective To investigate the effect of Paris polyphylla on tumor growth and life time of H22 tumorbearing mice. Methods Mice were modeled by injecting 0.2 m L H22 tumor cells subcutaneously.Solid tumor mice were divided into model group（ gastric perfusion of 20 m L / kg distilled water,once a day）,positive group（ intraperitoneal injection of 40 mg / kg cyclophosphamide,once every other day）,high-dose group（ 2. 4 g / kg）（ intragastric administration of Paris polyphylla）,middle-dose group（ 1.2 g / kg） and low-dose group（ 0.6 g / kg）（ intragastric administration of Paris polyphylla,once a day）,30 cases in each. After successive administration for 14 days,tumor tissues were cut to weigh and tumor inhibiting rates were calculated. The experiment was also conducted in ascetic tumor-bearing mice with same administration.Life time of all was recorded to calculate life extension rate. Results Compared with model group,weights of tumor in positive group and 3 dose-groups all reduced,the difference was significant（ P〈0.01）; while the weight reduced more in high-dose group than in middle-and low-dose groups,the differences were also significant（ P〈 0.05 or P 〈 0. 01）. Compared with model group,life time in other 4 groups all prolonged,especially in the high-dose group（ P 〈 0. 05 or P 〈 0. 01）. Conclusion Paris polyphylla has a dramatic tumor inhibitory effect in H22tumor-bearing mice and can prolong their life time.
