摘 要:目的探索DNA条形码技术在蝙蝠种类鉴定中的应用。方法基于kimura双参数模型计算23只蝙蝠Cyt B序列的种内种间遗传距离。结果种间遗传距离显著大于种内遗传距离,通过NJ法构建系统发育树聚类分析,不同种类的蝙蝠位于不同的分支,同属的蝙蝠分支较不同属的蝙蝠分支较短,种内分支则更短。种间遗传距离均在1.3以上,种内遗传距离均在0.21以下,与NCBI序列比对结果一致,与形态学鉴定结果一致(20只菲菊头蝠,1只马铁菊头蝠,1只大卫鼠耳蝠,1只小黄蝠)。结论 DNA条形码技术对于本研究中所涉及的蝙蝠种类具有较好的区分效果,可以作为一种有效的工具在蝙蝠鉴定中进行应用。Objective To explore the application of DNA barcoding in the identification of bats. Methods The CytB genes were amplified and sequenced from 23 bats samples collected in Field of Southeast China. The intraspecific and interspecific genetic distances were calculated with kimura - 2 - parameters model. Results The interspecific genetic distances (over 1.3 ) was significantly greater than intraspeeific genetic distances (less than 0. 21). Different species were in different branches, the branch in the same genus was shorter than the branch in different genera, and the branches in intraspecies were even shorter. Conclusion The species of bats can be well distinguished with CytB bareodes, and DNA bareoding can be effectively used to identify bat species identification. The 23 bats had the same classification results with morphology and CytB bareodes (Rhinolophus pusillus, Rhinolophus ferrumequinum, David Myotis, yellow bat).
