摘  要:目的探讨超极化激活环核苷酸门控阳离子通道2蛋白(HCN2)和细胞膜锚蛋白(ANK2)在心源性猝死(SCD)病人窦房结的表达水平。方法收集因心源性疾病猝死的个体心脏14例作为SCD组;选择无心脏疾病的死亡个体心脏标本13例作为非SCD组。准确切取心脏窦房结组织,石蜡包埋,部分HE染色,采用免疫组织化学的方法分别测定两组HCN2和ANK2蛋白表达水平。结果 SCD组HCN2在窦房结组织中呈棕黄色颗粒,非均质分布在核旁;SCD组ANK2稀疏分布于窦房结细胞浆内。同非SCD组相比,SCD组HCN2蛋白表达水平上调和ANK2蛋白表达水平下调(0.29±0.03vs 0.16±0.07;0.09±0.06vs 0.23±0.06,P均<0.05)。结论 SCD患者窦房结HCN2基因表达水平上调,ANK2基因表达水平下调。Objective To investigate the expression level of hyperpolarization-activated cyclic nucleotide-gated channel 2(HCN2) and ankyrin-B (ANK2) in sinoatrial node in patients with sudden cardiac death(SCD). Meth- ods We collected 14 cases anatomical specimens of sudden death due to cardiac disease as the SCD group and 13 ca- ses anatomical specimens without cardiac disease as the non-SCD group. We accurately cutted cardiac sinus node knot tissue, after paraffin embedded, partial sections were dyed with H^E staining, partial were estimated the ex- pression level of HCN2 and ANK2 proteins by using immunohistochemical method in the two groups respectively. Result Compared to non-SCD group, the expression level of HCN2 was increased and the expression level of ANK2 protein was decreased in SCD group (0.29±0.03 vs 0.16±0.07;0.09±0.06 vs 0.23±0.06,all P〈0.05). Conclusion The expression level of HCN2 is increased and the level of ANK2 protein is decreased in SCD patients.
