摘  要:为了确定咪达唑仑对中枢神经神经元细胞氨基酸类递质谷氨酸(Glu)及甘氨酸(Gly)含量的影响,试验以胎鼠为研究动物进行神经元的体外培养,分别设置高、中、低(60,5,3.5μg/m L)3个药物浓度,每个药物浓度设3组平行试验,然后采用ELISA法检测样品中Glu与Gly的含量,对比含量变化分析咪达唑仑对大鼠神经元氨基酸类神经递质的影响。结果表明:不同浓度咪达唑仑对Glu呈现先增强后抑制的作用,而对Gly的作用表现为先抑制后增强。说明不同浓度咪达唑仑在麻醉过程中抑制兴奋类氨基酸神经递质,增强抑制类氨基酸神经递质。The aim of this study is to determine the effects of midazolam on amino acid neurotransmitters glutamate ( Glu ) and glycine (Gly) content in central nervous system neurons. The experiment takes fetal mouse as the research animal to cultured neurons in vitro, and set 60μg/mL,5 μg/mL, 3.5 μg/mL high, medium, low three different drug concentrations. Three parallel experiments were set up for each drug concentration. Then using ELISA to detect the content of Glu and Gly in the sample, and compare the content variations to analyze the effect of midazolam on the amino acid neurotransmitter in rat neurons. The experimental results show that the effect of different concentrations of midazolam on Glu was firstly enhanced and then inhibited, but the effect on Gly was firstly inhibited and then enhanced. As can be seen, different concentrations of midazolam in anesthesia process inhibit excitatory amino acid neurotransmitter, and enhance inhibited amino acid neurotrans- mitter.
