摘 要:目的研究瘤果黑种草子挥发油(volatile oil fromNigella glandulifera,VONG)对小鼠单核/巨噬细胞RAW264.7及豚鼠气管平滑肌的作用。方法细胞水平实验:分为空白对照组、脂多糖(LPS)组、水煎液组(100μg/mL)和VONG 6.25、12.5、25μg/mL组,采用酶联免疫吸附法(ELISA)检测VONG对LPS刺激的小鼠单核/巨噬细胞RAW264.7分泌白介素-8(Interleukin-8,IL-8)、白介素-6(Interleukin-6,IL-6)的影响,采用Griess Reagent法检测RAW264.7细胞上清液中一氧化氮(nitric oxide,NO)的浓度;动物水平实验,分为生理盐水组、阳性对照组(氨茶碱0.008g/L)、水煎液组(10g/L)和VONG 3、10、30g/L组,采用离体气管恒温灌流的方法,制备豚鼠气管螺旋条标本,通过观察其张力变化考察药物对乙酰胆碱(acetylcholine chloride,Ach)和组胺(histamine,His)所致痉挛离体豚鼠气管平滑肌的解痉作用。结果与LPS组比较,VONG 6.25、12.5、25μg/mL组RAW264.7细胞IL-6、IL-8及NO水平降低(P<0.05),且呈一定的量效关系;VONG 3、10、30g/L具有可明显地舒张Ach及His引起的豚鼠支气管平滑肌痉挛,差异具有统计学意义(P<0.05)。结论 VONG具有抑制小鼠单核/巨噬细胞RAW264.7分泌IL-6、IL-8及NO及支气管解痉功能。Objective To study the effect of volatile oil fromNigella glandulifera(VONG)on RAW264.7cells and guinea pig tracheal smooth muscle.Methods In cellular level experiments,RAW264.7cells were divided into blank control group,LPS group,water decoction group(100g/mL)and VONG 6.25g/mL group,VONG 12.5g/mL group,VONG 25g/mL group.Interleukin-8(IL-8),Interleukin-6(IL-6)were detected by enzyme-linked immunosorbent(ELISA)Nitric oxide in the supernatant fluid(NO)concentration were detected by Griess Reagent method.In the animal experiments,the guinea pig were divided into normal saline group,positive control group(aminophylline 0.008g/L),water decoction group(10g/L),VONG 3g/L group,VONG 10g/L,VONG 30g/L groups.The effects of different concentration(3g/L,10g/L,30g/L)of VONG and water decoction fraction on spasm tracheal smooth muscle caused by histamine and acetylcholine in guinea were observed.Results Compared with the model groupcompared,the different concentration(6.25,12.5,25μg/mL)of VONG can inhibit the excessive secretion of,IL-6,IL-8and NO in RAW264.7cells,and there was a dose effect relationship(P〈0.05).The different concentration(3g/L,10g/L,30g/L)of VONG could reduce the tension of guinea pig bronchus smooth muscle contraction induced by acetylcholine chloride(Ach)and histamine(His)(P〈0.05)significantly.Conclu-sion The VONG could inhibit secretion of monocyte/macrophage RAW264.7mice IL-6,IL-8and NO and bronchodilation effects.
