摘  要:目的探讨贝母素乙对人胃癌BGC823细胞的化疗增敏作用及其相关机制。方法采用MTT、结晶紫染色法观察贝母素乙与阿霉素联用对BGC823细胞的生长抑制作用。Western blotting法检测贝母素乙与阿霉素联用作用于BGC823细胞后增殖相关蛋白的表达。结果贝母素乙与阿霉素联合应用可使BGC823细胞的抑制率增加(P<0.05),阿霉素的细胞毒性增加,化疗药物的IC50值降低(P<0.01),明显影响BGC823细胞的增殖。Western blotting结果显示,CyclinD1、P-AKT的表达均下调。结论贝母素乙可增加BGC823细胞对阿霉素的敏感性,降低P-AKT、CyclinD1的表达。Objective To research the effects and its mechanism of peimininetoenhance the chemotherapeutic sensitivity of human gastric cancer cell BGC823.Methods Using doxorubicine to treat BGC823 cell with or without peiminine,the effects of growth inhibition were determinined by MTT assay and crystal violet staining assay.The expression of growth inhibition proteins were detected by western blotting when peiminine treatment combined with doxorubicine in BGC823 cell.Results Compared with using doxorubicin only,doxorubicin in combination with peiminine increased the inhibition rate(P〈0.05),decreased the value of IC50(P〈0.01),enhanced the cytotoxicity of doxorubicin and inhibited proliferation obviously in BGC823 cell line.The expression of CyclinD1,P-AKT were down regulated by pmiminine combined with doxorubicin.Conclusion Peiminine enhances doxorubicine′s Chemotherapy sensitivity,and the expression of P-AKT,CyclinD1 were decreased in human gastric cancer cell BGC823.
