摘 要:目的采用超高效液相色谱-四级杆串联飞行时间质谱法(UPLC-Q-TOF-MS)对亚香棒虫草提取物的主要化学成分进行分析鉴定。方法采用Agilent SB-C18(50 mm×4.6 mm,1.8μm)色谱柱,乙腈-水(0.1%甲酸)溶液为流动相梯度洗脱,流速0.25 m L/min,柱温30℃;信息关联模式(IDA)正、负离子分别采集。结果结合Peak View软件的Formula Finder等功能、数据库及二级碎片裂解规律,共鉴定出26个化合物,其中有9个为虫草中未曾报道物质,且含2个未知化合物;其主要化学成分包括生物碱、糖苷、核苷、氨基酸等成分。结论该方法精确、可靠、高效,适用于亚香棒虫草成分的快速鉴定,为亚相棒虫草的开发利用以及阐明其药效物质基础提供参考。Objective To analyze and identify the main chemical constituents of extract from Cordyceps hawkesii by using method of ultra performance liquid chromatography-qudropole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Methods UPLC condition was as the following: Agilent SB-C18 (50 mm ×4.6 mm, 1.8 μm) column with gradient elution consisting of0.1% formic acid in water and acetonitrile, at a flow rate of 0.25 mL/min. The column temperature was set at 30℃. MS analysis was based on information associated mode (IDA) that positive and negative ions were respectively collected. Results Mass spectrometry combined with "Formula Finder" of PeakView software and database, a total 26 compounds were identified as alkaloids, glycosides, nucleosides, and amino acids. 9 of them had never been reported in Cordyceps hawkesii, and 2 of them were known. Conclusion The method is accurate, reliable and efficient for rapid identification of compounds in Cordyceps hawkesii. The results provide references for the development and utilization of Cordyceps hawkesii, as well as finding out its phar- macodynamic material basis.
