摘  要:长链非编码RNA为长度大于200nt的非编码RNA,具有组织表达特异性,且表达丰度较mRNA低。在前期肉牛肌肉和脂肪组织的lncRNA组学测序分析中筛查并获得一个长度为755nt的差异表达长链非编码RNA,将其命名为lncRNA_(TCONS-585)。经基因组序列比对,该novel lncRNA_(TCONS-585)序列位于牛第5号染色体MCHR1和PMP34两个基因之间,由两个外显子构成。对lncRNA_(TCONS-585)结构分析后通过RT-PCR验证了lncRNA_(TCONS-585)在靶向组织中的表达水平以及在不同组织中的表达谱。结果表明,lncRNA_(TCONS-585)在牛的不同组织中表达量具有较大差异,其在肌肉组织中表达水平最高,同时在白色脂肪和棕色脂肪组织当中也有表达,且表达量显著高于肝、肺和肾。同时,我们发现干扰lncRNA_(TCONS-585)可降低其邻近基因MCHR1的表达量。因此lncRNA_(TCONS-585)可能作为调控因子通过介导靶基因参与肉牛生长发育和能量代谢过程。LncRNA refers to a kind of non coding RNAs containing more than 200 nucleotides. The expression abundance of LncRNA is lower than normal mRNA with tissue-specific expression. In our previous study,a novel 755 nt LncRNA named lncRNA discovered in bovine white adipose tissue using RNA-seq. Genome alignment showes that lncRNA is encoded by 2 ex- ons,locating between MCHR1 and PMP34 in chromosome 5. In order to verify the existence of In cRNAand collect expression profile in different tissues,RT-qPCR was used in this study. Results showed that lncRNA existed in several tissues with different expression level and highly expressed in heart and muscle;besides,the expression of lncRNA in fat tissue was significantly higher than liver,lung and kidney. Interestingly, the expression of MCHR1, the con- tiguous gene of lncRNA, was significantly decreased when lncRNAR had been knocked-down by RNAi. Thus,lneRNA may take part in growing development and energy metabolism in cattle as a regulatory factor of MCHR1.
