摘  要:采用PCR-SSCP技术对45头种公牛的ACTB(β肌动蛋白)基因的第4外显子的单核甘酸多态性进行了检测。结果显示:在ACTB基因第4外显子上存在2种基因型,即AA型和AB型。对该多肽片段克隆测序,结果在ACTB基因第4外显子上存在G378C突变。用最小二乘法分析ACTB基因的第4外显子不同基因型与精液品质的相关性,发现ACTB基因第4外显子AA型精子浓度、鲜精畸形率以及冻精畸形率显著高于AB型(P<0.05),AA型鲜精活力、鲜精活精子比率极显著高于AB型(P<0.01),AA型冻精活精子比率显著低于AB型(P<0.05)。The single nucleotide polymorphism of the ACTB gene in cattle was analyzed by PCR- SSCP. The results showed that two genotypes, AA and AB were detected in exon 4 of ACTB gene. The polymorphism fragments were cloned and sequenced. The sequencing results indicted there was one single nucleotide mutations:G378C in exon 4 of ACTB gene. The genetic correlations were obtained between genotypes of the exon 4 of ACTB gene and semen quality in cattles by the least square analysis. The results indicated that different genotypes of ACTB in exon 4 significantly af- fected semen quality. Bulls with the AA genotype had a significantly higher sperm concentration and a significantly higher percentage of deformity in fresh and frozen semen than those of the AB genotypes (P〈0.05). Further,bulls with the AA genotypes showed an significantly lower per- centage of living semen (P〈0.01) and a significantly lower percentage of acrosome integrity (P〈 0.05) in fresh than those of the AB genotypes.
