新疆乌恰县人体泡球蚴线粒体DNA分析

摘　　要：目的通过线粒体DNA测序初步确定新疆乌恰县多房棘球蚴的来源。方法新疆维吾尔自治区南疆乌恰县泡球蚴病患者5例,采集其病灶组织,提取DNA,PCR扩增多房棘球绦虫线粒体DNA Nad2序列,并与已发表的泡球蚴线粒体DNA Nad2序列进行序列比对和系统进化分析。结果新疆南疆乌恰县是一个新的泡型包虫病流行区。Nad2基因序列比对分析及遗传距离矩阵显示,5例AE患者病灶扩增的Em Nad2基因存在差异,人体感染的虫株之间存在低的遗传变异。Nad2基因系统进化树分析表明,这些虫株是亚洲多房棘球绦虫亚型,包含3个单倍体型。结论新疆乌恰县多房棘球绦虫流行株属于亚洲基因型,至少有3种单倍体型。关于乌恰县多房棘球绦虫终末宿主和中间宿主的种群以及其在疾病的传播中的作用需要进一步证实。Objectives Alveolar echinococcosis （AE） is a life-threatening human parasitic disease caused by Echinococcus multilocularis, which is transmitted between small mammals and dogs or foxes. The aim of this study was to identify the genetic variation of E. multilocularis isolates collected from patients with AE in Wuqia County in southern Xinjiang. Methods Five cases of AE in Wuqia County were verified based on surgical specimens and a pathological examination. The prevalence of AE in the county is 13.2/100 000. DNA was extracted from liver tissue removed surgically from patients with AE. A mitochondrial Nad2 fragment was amplified and sequenced. Sequence alignment and phylogenetic analysis were performed to compare the genetic variation of the gene sequences with homologous sequences from the GenBank databases. Results Wuqia County is an area with new outbreaks of AE. The Nad2 gene of E. rnultilocularis differed in the five patients with AE. Three haplotypes were identified among these 5 isolates. Comparison indicated that these isolates are an Asian type of E. multilocularis. Conclusion Wuqia County is an area with outbreaks of AE. However, the lifecycle of E. multilocularis （which includes the role of small mammals and dogs in the transmission of AE） needs to be determined further.
