摘 要:为提高布莱凯特黑牛肉质品质,参照NCBI公布的牛PID1基因mRNA序列设计引物,以117头布莱凯特黑牛为试验材料,提取其眼肌总RNA,通过RT-PCR对布莱凯特黑牛PID1基因cDNA进行克隆测序,对PID1基因的每个外显子进行SNP检测,并分析其多态性。结果显示:布莱凯特黑牛PID1基因cDNA全长2 561bp,包括1个612bp的完整开放阅读框,编码203个氨基酸;在第2外显子上存在2处变异位点,但仅1处位点的突变引起氨基酸序列变化,表明PID1基因在种内保守性较强。For exploring BLKT black cattle PID1 (Phosphotyrosine interaction domain containing 1) gene to serve as a molecular marker with breeding, in this paper, the authors referenced cattle PID1 gene mRNA sequence from the NCBI and designed primers. Then, total RNA of BLKT black cattle was extracted, and the PID1 gene cDNA was cloned by RT-PCR. Prokaryotic expression and bioinformatics analysis were conducted. Results. Black cattle PID1 gene cDNA was 2561 bp, including a complete open reading frame of 612 bp, which coded 203 amino acids. By studying 117 BLKT black cattle, the SNP detection was executing on each exon of PID1 gene. Only two mutation loci exists in the exon 2, but only one mutation causes the change of the amino acid sequence. Conclusion. PID1 gene is conservative in interspecies.
