摘 要:该文以电致化学发光(ECL)物质鲁米诺作为还原剂制备了鲁米诺功能化的金银纳米颗粒(LumAu Ag NPs),并将其成功固载到二氧化锰纳米颗粒(MnO_2NPs)的表面,制备了一种新的纳米复合材料LumAu Ag@MnO_2。该复合材料具有较低的生物毒性,极大的比表面积和良好的电催化能力,可以用作蛋白质的纳米载体构建生物传感器。文中以该材料为二抗(Ab2)载体制备了一个能够高灵敏检测阿兹海默症标志物β淀粉样蛋白(Aβ)的夹心型ECL免疫传感器。由于Lum-Au Ag@MnO_2具有良好的导电性和过氧化氢催化活性,能够有效提高luminol-H_2O_2体系的电化学发光效率,进而检测到较强的ECL信号。实验表明,该传感器在β淀粉样蛋白抗原的测定上有良好的线性关系,线性范围为100 ng/m L^1 pg/m L,最低检测限为0.33pg/m L。该传感器对Aβ的检测具有灵敏度高、检测范围宽、制作简便、成本低的优点,并且具有良好的选择性和重现性。In this work,the electrochemiluminescence(ECL) luminophore of luminol was utilized as reductant to synthesize luminol functionalized gold-silver nanoparticles(Lum-Au Ag NPS).And the Lum-Au Ag NPs were immobilized on the surface of MnO_2 NPs with low bio-toxicity,large specific surface area,and favourable chemical catalytic activity(Lum-Au Ag@MnO_2),which could be used as protein carrier to fabricate biosensors.Encouraged by the advantages above,a sandwiched electrochemiluminescence(ECL) immunosensor based on Lum-Au Ag@MnO_2 was constracted for ultrasensitive detection of β-amyloid protein(Aβ,a biomarker for Alzheimer's disease).LumAu Ag@MnO_2 exhibited excellent conductivity and admirable catalytic activity in H_2O_2 decomposition,which could increase the ECL efficiency of luminol-H_2O_2 system and generate strong ECL signal.The experiment proved that the immunosensor had good linear range of 100 ng/m L to 1 pg/m L and an ultralow detection limit of 0.33 pg/m L in the determination of β-amyloid protein,exhibiting high sensitivity and controllable reaction in Aβ detection.
