摘 要:目的:观察小半夏加茯苓汤醇提物对胃癌细胞BGC-823的凋亡作用以及诱导其凋亡发生的机制。方法:运用MTT法检测不同浓度的小半夏加茯苓汤醇提物对BGC-823的增殖抑制作用并计算抑制率;以罗丹明123为细胞染色剂,采用流式细胞术检测线粒体跨膜电位的改变;实时荧光定量PCR检测Bax、Bcl-2和Cyt-c表达的变化。结果:MTT结果显示小半夏加茯苓汤醇提物对BGC-823具有抑制作用,随着给药浓度的增加和作用时间的延长,抑制率逐渐升高,细胞活力逐渐减弱,与空白对照组比较有显著统计学差异(P<0.01)。流式细胞术结果显示,与空白对照组相比,小半夏加茯苓汤醇提物能够显著降低线粒体跨膜电位(P<0.01);qRT-PCR结果显示,Bax基因表达显著升高(P<0.01),Cyt-c基因表达升高(P<0.05),Bcl-2基因的表达降低(P<0.05)。结论:小半夏加茯苓汤醇提物可以抑制BGC-823细胞活力,诱导细胞凋亡并减小Bcl-2/Bax基因表达比例,增强Cyt-c基因表达,其机制可能与线粒体为核心的凋亡通路相关。Objective To observe the effect of Xiao Banxia Plus Fuling Ethanol Extract on gastric cancer cell BGC - 823 apoptosis and its mechanism induced apoptosis. Methods MTT was used to detect different concentrations of Xiao Banxia Plus Fuling Ethanol Extract on proliferation of BGC - 823 inhibition and calculate the inhibition rate ; Rhodamine 123 cell stain, Flow Cytometry was used to detect changes of mitocbondrial membrane potential; Real - time PCR to detect Bax, Bcl - 2 and Cyt - c the change of expression. Results MTT results showed that Xiao Banxia Plus Fuling Ethanol Extract has an inhibitol7 effect on BGC -823, with the increase of drug concentration and action time, the inhibition rate increased gradually, the cell viability decreased, there wassignificant difference- compared with the control group (P 〈 0. 01 ) . Flow Cytometryresuhs showed that, compared with the control group, Xiao Banxia Plus Fuling Ethanol Extract cansignificantlyreduce the mitoehondrial membrane potential (P 〈 0. 01 ) ; qRT- PCR results showed that bax- geneexpressionincreased significantly, Cyt - c gene expressionincreased, the Bcl - 2 gene decreased ( P 〈 0. 05 ) . Conclusion Xiao Banxia Plus Fuling Ethanol Extract could reduce the BGC - 823 ceils viability, induce apoptosis and down - regulate the proportion of gene expression of Bcl -2, Ban in BGC -823 cells, Enhance Cyt -c gene expression. The mechanism may be related to the mitochon- drial apoptotic pathway.
