摘  要:病毒是导致甘薯产量降低和品质改变的主要因素,生产上主要采用茎尖脱毒的方法解决这一问题,但茎尖脱毒的效果主要采取指示植物法、酶联免疫吸附法进行测定,这些方法各有优缺点,因而在生产上没有得到广泛应用。随着现代分子生物学技术的发展,反转录聚合酶链式反应技术在甘薯病毒检测上的应用越来越广泛。采用RT-PCR技术可检测出甘薯组织中含量极低的病毒RNA,具有灵敏度高、特异性强等优点。本研究综述了RT-PCR检测技术的原理、分类、操作中存在的问题、解决办法及其在甘薯病毒检测中的应用情况,旨在为从事甘薯病毒检测技术的科研工作者提供一些思路和方法。Virus is the key factor which can make sweet potato yield decreased and the quality losses, in order to resolve this question, the method of stem tip detoxification is used in sweet potato production. The detection on effect of stem tip detoxification can be finished by three ways such as Indicator plant method, antiserum assay method and enzyme linked immunosorbent assay. These three ways are not widely used in the production because each has advantages and disadvantages. With the development of modern molecular biological technology, reverse transcription-polymerase chain reaction has been widely used in detection of Sweet potato viruses. Little RNA viruses from Sweet potato tissue can be detected by RT-PCR which are with such advantage as more sensitive and specificity. In order to give good guidance for those who are engaged in detecting sweet potato viruses, principle and classification about RT-PCR, existing problem in the course of operation and corresponding measure to deal with them and its application in detecting sweet potato viruses are summarized in this paper.
