摘 要:为了优化红掌的离体再生条件,本研究以红掌"粉冠军"无菌苗的根尖为外植体,对愈伤诱导、不定芽再生及植株生根过程中植物生长调节剂的浓度进行优化,同时通过半薄切片技术对愈伤发生和分化过程进行组织学观察。试验结果表明,"粉冠军"根尖置于培养基1/2 MS,添加0.2 mg/L噻苯隆(TDZ)和30 g/L蔗糖,黑暗下培养2个月后,外植体出愈率达到86%。将愈伤组织转接到MS基本培养基,同时添加1.0 mg/L吡效隆(CPPU)和25 g/L蔗糖,16 h/8 h光照周期条件下不定芽的分化率达到100%,发生数量平均为6.6个/块。将不定芽转接于添加0.5 mg/L吲哚丁酸(IBA)、0.4 g/L活性炭和25 g/L蔗糖的MS培养基上,根系发育良好。组织切片观察结果表明,愈伤起源于根尖分生组织,而不定芽起源于愈伤表面组织。研究建立了红掌"粉冠军"的一种离体再生方法,并为红掌愈伤组织的形态发生研究提供参考。In order to optimize the culture condition for in vitro regeneration of Anthurium andraeanum, root-tips cut from in vitro plantlets of Anthurium andraeanum "pink champion"were used as explants, the concentration of plant growth regulators in callus induction, buds differentiation and rooting were examined and optimized, along with surveying the process of callus induction and differentiation by semi-thin histological section. As the results,when root-tips of"pink champion"were cultured on 1/2 MS medium containing 0.2 mg/L thidiazuron(TDZ) and30 g/L sucrose in darkness after two months, the percentage of callus induction from explants was 86%. The highest percentage(100%) and number of buds(6.6 per callus-mass) regenerated from callus were recorded on MS medium supplemented with 1.0 mg/L forchlorfenuron(CPPU) and 25 g/L sucrose in 16 h/8 h photoperiod. Buds were transferred onto MS medium supplemented with 0.5 mg/L indolebutyric acid(IBA), 0.4 g/L active charcoal and25 g/L sucrose. Roots were induced from base of buds with well growth. By using semi-thin histological section, it was suggested that callus were originated from the meristematic tissue of root tip, and the buds were derived from surface tissue of callus. We established an optimized method for in vitro regeneration of Anthurium andraeanum"pink champion", and these results wound benefit the morphogenetic study of callus in Anthurium andraeanum.
