HPLC法测定一匹绸中东莨菪苷和东莨菪内酯的含量

摘　　要：建立了一匹绸中东莨菪苷和东莨菪内酯的含量测定方法。色谱柱为ODS-C18,用甲醇-0.2%磷酸梯度洗脱,流速为0.8 m L/min,柱温30℃,检测波长为346 nm。东莨菪苷、东莨菪内酯0.019 2~0.096 0mg/m L(r=0.999 9)、0.004 48~0.022 40 mg/m L(r=0.999 5)呈现良好的线性关系,方法学验证结果符合要求。本方法简便准确,精密度和重复性好,可作为一匹绸药材的含量检测方法。A HPLC method was established for determination contents of Scopolin and Scopoletin in Argyreia acuta Lour. The separation was performed on ODS-C18 column,with gradient elution in methanol-0.2% H3PO4 solution at the flow rate of 0.8m L/min. The column temperature was 30 ℃,and the detection wavelength was 346 nm. The results showed that Scopolin and Scopoletin had good linear relationships in range of 0.019 2~0.096 0 mg/m L（r=0.999 9） and 0.004 48~0.022 40 mg/m L（r=0.999 5）. The methodological results were in line with requirements. The method is simple and accurate, with good precision and repeatability, which could be used for the quality control of Argyreia acuta Lour.
