摘 要:目的探讨羊膜细胞体外诱导人脂肪干细胞向表皮方向分化的可能性。方法体外分离培养人脂肪来源的干细胞(human adipose-derived stem cells,hADSCs),将P3代hADSCs与人羊膜细胞(human amniotic cells,HACs)共培养,培养液中添加EGF,光镜下观察hADSCs细胞形态变化,并对细胞进行鉴定。结果随时间延续,hADSCs细胞数量逐渐减少,同时细胞突起逐渐变短,胞体由长梭形逐渐变为上皮样,21d后,经免疫荧光染色鉴定,发现所获细胞高表达CK19。结论通过人脂肪干细胞与人羊膜细胞共培养可获得CK19阳性的表皮干细胞。Objective To explore the possibility of inducing the differentiation of human adipose-derived stem cells (hADSCs) into epidermal cells by co-culture with aminionic cells in vitro. Methods HADSCs were isolated and cultured in vitro, the third passage of which were co-cultured with human amniotic cells (HACs) in a medium supplemented with epidermal growth factor ( EGF). The morphological change of hADSCs was observed by light microscopy, and the cells were identified by immunofluorescent staining. Re- suits The number of hADSCs decreased accompanied by cell processes shortened gradually with time, the long fusiform cells turned into epithelioid. After 14d of co-culture, CK19 were found over expression in these cells by immunofluorescent staining. Conclusion CK19- oositive eoidermal stem cells were obtained by co-culture hADSCs with HACs.
