摘  要:目的观察温肺降浊方对血管性痴呆(VD)大鼠的认知功能的改变及其对脑内相关血氧动力改变诱导的血管内皮生长因子(VEGF)以及一氧化氮合酶(iNOS)基因表达,以评价温肺降浊方的脑保护作用。方法实验大鼠随机分为:空白对照组、假手术组、模型组、尼莫地平组、温肺降浊方组,采用双侧颈总动脉永久结扎法制备VD动物模型,以Morris水迷宫试验和穿梭箱实验检测来评价大鼠的认知功能,采用免疫组化法测定大鼠VEGF和iNOS的表达。结果模型组的潜伏期最短,错误次数最多(P<0.01)。温肺降浊方组的潜伏期大于尼莫地平组(P<0.01)。温肺降浊方组及尼莫地平组潜伏期大于模型组(P<0.01)。同时,假手术组与空白对照组大鼠跨越平台次数、目标象限次数无差异(P>0.01);模型组跨越平台次数、目标象限次数均低于假手术组(P<0.01);温肺降浊方组及尼莫地平组越平台次数、目标象限次数高于模型组(P<0.01);温肺降浊方组与尼莫地平组指标比较,差异有统计学意义(P<0.05)。温肺降浊组、尼莫地平组VEGF阳性细胞多于模型组(P<0.01),温肺降浊方组阳性细胞数多于尼莫地平组(P<0.05)。各组大鼠海马区iNOS阳性细胞数分布状况:假手术组及空白对照组iNOS阳性细胞数低于模型组,温肺降浊方组和尼莫地平组iNOS阳性细胞数低于模型组,温肺降浊方组iNOS阳性细胞数与尼莫地平组比较,差异有统计学意义(P<0.05)。结论温肺降浊方能抑制血管性痴呆大鼠iNOS表达,上调VEGF的表达,改善认知功能。Objective To observe the changes in cognitive function in the rats of vascular dementia(VD) treated with wenfei jiangzuo formula and the expressions of VEGF and iNOS induced by the changes of relevant blood oxygen indexes so as to evaluate the brain protective function of wenfei jiangzhuo formula.Methods The experimental rats were randomized into a blank control group,a sham-operation group,a model group,a western medicine control group and a wenfei jiangzhuo formula group. The permanent ligation of bilateral common carotid artery was used to prepare VD animal model. Morris water maze test and shuttle box test were used to evaluate the cognitive function of rats. IHC assay was used to determine the expressions of VEGF and iNOS. Results In the model group,the latent duration was the shortest and the mistake frequency was the highest(P〈0. 01). The latent duration in the wenfei jiangzhuo formula group was longer than that in the nimodipine group(P〈0. 01). The latent duration in the wenfei jiangzhuo formula group and the nimodipine group was all longer than that in the model group(P〈0. 01). The differences in the frequency of step-over platform and target quadrant were not significant between the sham-operation group and the blank control group(P〈0. 01). The frequency of step-over platform and target quadrant in the model group was lower than that in the sham-operation group(P〈0. 01). The indexes were different significantly between the wenfei jiangzhuo formula group and the nimodipine group(P〈0. 05). The VEGF positive cells in the wenfei jiangzhuo formula group and the nimodipine group were more than those in the model group(P0.01).The positive cells in the wenfei jiangzhuo formula group were more than those in the nimodipine group(P0.05).Regarding the distribution of iN OS positive cells in the hippocampus area,iN OS positive cells in the sham-operation group and the blank control group were lower than those in the model group,the results in the wenfei jiangzhuo formu
