补肾填精中药对妊娠期小鼠子宫内膜容受性影响的研究

摘　　要：目的观察补肾填精中药对妊娠小鼠子宫内膜容受性的影响。方法 120雌性小鼠随机分为治疗组和对照组,治疗组予调经助孕胶囊,对照组予六味地黄丸灌胃2周,动情期小鼠按雌雄2∶1比例合笼,次晨阴道查有阴栓为妊娠第1天。两组分别于妊娠第3、5、7天,取每组20只妊娠小鼠处死,取出子宫,刮取内膜。测定内膜αvβ3 mRNA及蛋白的表达。观察第5天妊娠小鼠胚泡着床数。结果整合素β3在治疗组和对照组早孕小鼠子宫内膜中的表达随妊娠天数呈规律变化,在治疗组的表达高于对照组,差异有统计学意义(P<0.05)。妊娠第5天治疗组孕鼠胚泡着床数目明显多于对照组,差异有统计学意义(P<0.05)。结论补肾填精中药通过增加小鼠子宫内膜整合素β3的表达,提高胚泡着床数目,而改善子宫内膜容受性,提高妊娠率。Objective To observe the impacts on endometrial receptivity in the pregnant mice treated with herbal medicine for tonifying the kidney and filling-up essence. Methods One hundred and twenty female mice were randomized into a treatment group and a control group. Lavage with tiaojing zhuyun capsules was used in the treatment group and the lavage with liuwei dihuang wan in the control group for 2weeks. The mice at estrum stage were fed in the same cage at the ratio of 2 to 1 of the females and males. In the morning on the second day,the vaginal plug was examined as the 1stday of pregnancy. Separately,on the3 rd,5thand 7thdays of pregnancy,20 pregnancy mice were sacrificed in each group,the uterus was collected and the endometrium was scraped. The expression of αvβ3 mRNA and its protein was determined. On the 5th day of pregnancy,the implanted blastocyst count was observed. Results The integrin β3 expression changed regularly in duration of pregnancy in the Chinese medicine group and control group. The expression in the Chinese medicine group was higher than that in the control group,indicating the significant difference. On the5 th day of pregnancy,the implanted blastocyst count in the treatment group was apparently more than that in the control group,indicating the significant difference. Conclusion The herbal medicine for tonifying the kidney and filling-up essence improves endometrial receptivity and pregnancy rate through increasing the expression of endometrial integrin β3 and implanted blastocyst count in the mice.
