摘 要:目的:探讨药用爬山虎对牛II型胶原诱导性关节炎大鼠(CIA)关节滑膜细胞核周因子-κB(NF-κB)表达及血清IL-17水平的影响。方法:将40只6周龄雌性wistar大鼠随机分为空白对照组、模型对照组、雷公藤多苷对照组和爬山虎治疗组各10只。除空白对照组外,其余各组建立CIA模型,造模第7天开始予爬山虎汤剂和雷公藤水溶液灌胃治疗,连续用药4周,空白对照组和模型对照组灌服等量生理盐水,于末次给药2h后采集血清,处死大鼠,摘取双后肢膝关节分离滑膜,采用ELISA检测血清IL-17的表达,Western-blot检测关节滑膜NF-κB/P65的蛋白表达。结果:与空白对照组比较,模型组大鼠血清IL-17表达增加,爬山虎和雷公藤能够明显降低IL-17的表达(P<0.05);模型对照组大鼠关节滑膜细胞NF-κB/P65蛋白水平明显高于空白对照组(P<0.05),爬山虎与雷公藤多苷治疗组均能降低CIA大鼠关节滑膜细胞NF-κB/P65的表达,两组无显著性差异(P>0.05)。结论:药用爬山虎能够降低CIA模型大鼠血清IL-17的表达,下调关节滑膜细胞NF-κB的蛋白水平。Objective:To study the influence of medicinal parthenocissus on the expression of serum levels IL- 17 and synovial cells NF-κB in collagen -induced rats with rheumatoid arthritis. Method: 40 6 -week -old female Wistar rats randomly divided into a blank group, a model control group, a tripterygium glucosides treatment group and a medicinal parthenocissus treatment group. CIA models were established in all groups except the blank control group. After 7 days of modeling, lavage with Parthenocissus Decoction and ripterygium glucosides were given for four weeks successively, while the blank control group and the model control group were administered the same amount of normal saline. The serum was collected after 2 hours of the last gavage treatment. Cut the knee joints of both back limbs and removed their synovial membrane after the rats were killed. ELISA was used to measure the expression levels of IL - 17, and Western - blot was used to detect the expression of synovial NF - kappa B/P65 protein. Results: Compared with the blank group, the expression of IL -17 in serum increased in the model group, but decreased significantly in the parthenocissus group and the tripterygium glucosides (P 〈 0.05). In the model control group, cells were significantly higher than those in the control group ( P the levels of NF - κB/P65 protein in synovial 〈 0.05 ). The expression of CIA in rat synovial cells of NF -κB/P65 decreased in the parthenocissus group and the tripterygium glucosides group. There was no significant differences between the two groups (P 〉 0. 05 ). Conclusion: Medicinal parthenocissus could reduce the expression of IL - 17 in serum of CIA model rats, and down regulate the protein level in synovial cells of NF - κB.
