摘  要:目的观察针药并用对抑郁症睡眠障碍大鼠下丘脑、杏仁核、中缝背核5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)含量及其比值的影响。方法将40只SD大鼠随机分为正常组8只和造模组32只。正常组大鼠采用常规合笼饲养,不接受任何刺激与处置。造模组大鼠进行为期21 d的孤养+CUMS法+REM睡眠剥夺法造模,其中28只顺利完成造模,随机分为模型组、针刺组、药物组和针药组,每组7只。模型组仅造模不干预;针刺组取印堂、神庭配太冲进行针刺干预;药物组采用盐酸舍曲林片和阿普唑仑片的水溶液灌胃干预;针药组采用针刺配合水溶液灌胃干预。针刺组、药物组和针药组均每日治疗1次,连续干预7 d。比较各组大鼠下丘脑、杏仁核、中缝背核5-HT与5-HIAA含量及其比值。结果模型组造模后下丘脑、杏仁核和中缝背核的5-HT、5-HIAA含量较正常组出现明显下降(P<0.05)。针刺组、药物组和针药组治疗后下丘脑、杏仁核和中缝背核的5-HT、5-HIAA含量均较模型组出现了明显上调(P<0.05)。模型组下丘脑、中缝背核5-HIAA/5-HT比值与正常组比较,差异均具有统计学意义(P<0.05)。针刺组杏仁核及针药组下丘脑5-HIAA/5-HT比值与模型组比较,差异均具有统计学意义(P<0.05)。结论针药并用能对抑郁症睡眠障碍大鼠5-HT能神经系统的活跃程度起到一定的改善作用,能增加睡眠时间,提高睡眠质量。针药结合的疗效可能以针刺为主。Objective To investigate the effect of combined acupuncture and medicine on the 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) contents of hypothalami, amygdalae and dorsal raphe nuclei and their ratio in rats with sleep deprivation depression.Method Forty SD rats were randomized to a normal group of eight rats and a model making group of 32 rats. The normal group of rats was routinely fed together in one cage without any stimulation and treatment. The model making group of rats underwent 21 days' solitary feeding+ CUMS+ REM sleep deprivation. Twenty-eight rats with successful model making were randomized to model, acupuncture, medicine and combined acupuncture and medicine groups, seven each. The model group received model making and no intervention; the acupuncture group, intervention with acupuncture at Yintang, Shenting and Taichong; the medicine group, intervention by an oral gavage of sertraline hydrochloride tablets and alprazolam tablets in aqueous solution; the combined acupuncture and medicine group, intervention with acupuncture plus the aqueous solution. The three groups received seven consecutive days of intervention. The 5-HT and 5-HIAA contents of hypothalami, amygdalae and dorsal raphe nuclei and their ratios were compared between the groups.Result The 5-HT and 5-HIAA contents of hypothalami, amygdalae and dorsal raphe nuclei decreased significantly in the model group after model making compared with the normal group (P〈0.05) and increased significantly in the acupuncture, medicine and combined acupuncture and medicine groups after treatment compared with the model group (P〈0.05). There were statistically significant differences in the ratios between the 5-HIAA and 5-HT contents of hypothalami and dorsal raphe nuclei between the model and normal groups (P〈0.05). There was a statistically significant difference in the ratio of amygdalae 5-HIAA to 5-HT content between the acupuncture and model groups and in the ratio of hypothalamic 5-HIAA to 5-HT
