摘 要:目的研究糖尿病视网膜病变(DR)患者FOXO1基因表达改变,进一步了解DR的发病机制,为DR的治疗提供新的基因靶点和依据。方法收集眼科、内分泌科住院部及门诊糖尿病(DM)患者的临床资料,分为DR组110例,糖尿病正常眼底组(NDR)120例。以荧光定量PCR方法检测FOXO1 mRNA表达水平,用Western blot检测FOXO1蛋白表达水平。结果 DR组与NDR组相比,FOXO1 mRNA表达在DR组中显著升高,差异有统计学意义(P<0.05);DR组中FOXO1蛋白表达水平水平相比较NDR组明显增高(P<0.05)。结论 FOXO1在DR中的表达明显改变,这可能为DR提供潜在的治疗靶点。Objective The study the of forkhead box transcription factor O1 ( FOXO1 ) expression in patients with diabetic reti- nopathy ( DR), provide experiment evidences for further explore the mechanisms of DR developing and target therapy of for DR. Methods The glycosylated hemoglobin content were measured check -ups and ophthalmology with automatic biochemical analyzer;the protein and mRNA expressions of FOXO1 were detected by western blotting and real time polymerase chain reaction (RT- PCR). Results General data analysis presents that the course of the disease is longer in DR group. Compared with the NDR group, the mRNA and protein expressions of FOXO1 in DR were increased obviously (P 〈 0.05). Conclusion FOXO1 may taken part in the developing of DR, these found may provide an potential therapy target for DR.
