摘 要:目的:观察丁苯酞对脑缺血再灌注大鼠脑组织胰岛素样生长因子(IGF-1)表达的影响。方法:将50只SD大鼠随机分为假手术组(n=10)、脑缺血再灌注组(n=20)及丁苯酞治疗组(n=20),脑缺血再灌注组和丁苯酞治疗组又分为测体积组(n=10)及免疫组化组(n=10)。丁苯酞治疗组于模型制作前4d,每天给予丁苯酞胶囊,假手术组及脑缺血再灌注组正常喂养。建立大鼠局灶性脑缺血再灌注模型,缺血2h再灌注48h后进行神经功能缺损评分,免疫组化染色检测脑缺血半暗带的IGF-1表达,TTC染色检测脑梗死体积。结果:与假手术组相比较,脑缺血再灌注组及丁苯酞治疗组脑组织的IGF-1表达升高(P<0.05)。与脑缺血再灌注组相比较,丁苯酞治疗组大鼠神经功能缺损评分及脑梗死体积减少(P<0.05),大鼠脑组织IGF-1的表达升高(P<0.05)。结论:IGF-1是脑缺血再灌注损伤的保护性因子,丁苯酞可促进缺血再灌注大鼠脑组织中IGF-1的表达而发挥脑保护作用。Objective:To investigate the effect of butylphthalide on the expression of insulin-like growth factor-1(IGF-1)in brain tissue of rats with cerebral ischema-reperfuson.Methods:50SD rats were randomly divided into control group(n=10),cerebral ischema-reperfuson group(n=20)and butylphthalide treatment group(n=20),cerebral ischema reperfusion group and butylphthalide treatment group were assigned the volume test group(n=10)and the immunohistochemistry test group(n=10).The rats in butylphthalide treatment group were given butylphthalide everyday before 4days of MCAO.The rats in control group and cerebral ischemia-reperfusion group were given normal feeding.The focal cerebral ischemia-reperfusion model was established.The scores of neurological deficit were estimated in the rats at 2hafter ischema and 48 hafter reperfusion.Immunohistochemistry was used to detect the expression of IGF-1.TTC staining was used to investigate the infarct size.Results:Compared with control group,the expression of IGF-1in the brain tissue of rats in cerebral ischemia-eperfusion group and butylphthalide treatment group were increased(P〈0.05).Compared with cerebral ischemia-reperfusion group,the infarct size and the scores of neurological deficit in butylphthalide treatment group were decreased(P〈0.05),the expression of IGF-1in the brain tissue of rats in butylphthalide treatment group were increased(P〈0.05).Conclusion:IGF-1is a protective factor in cerebral ischemia reperfusion injury,butylphthalide could protect the brain injury through increasing the expression of IGF-1in brain tissue of rats with cerebral ischema-reperfuson.
