摘 要:目的:DNA甲基化转移酶(DNA methylation transferase,DNMT)表达异常可能在先天性心脏病(Congenital heart disease,CHD)心脏缺损中发挥重要作用。本文旨在探讨CHD儿童DNMT表达情况与非CHD儿童DNMT表达情况是否存在差异。方法:收集97例包含房间隔缺损(ASD)、室间隔缺损(VSD)或动脉导管未闭(PDA)的CHD儿童与50例非CHD儿童外周血,提取白细胞总RNA。采用及real-time RT-PCR检测两组儿童DNA甲基化转移酶1(DNMT1)及DNA甲基化转移酶3B(DNMT3B)mRNA表达水平。结果:CHD病例组儿童外周血中DNMT1和DNMT3BmRNA表达水平均较正常对照组高(P<0.01)。结论:DNA甲基化转移酶异常升高可能在CHD发病机制中发挥重要作用。Objective:The abnormal expression of DNA methylation transferase(DNMT)may play an important role in the congenital heart disease.This study is to explore the differences in expression of DNMT between the children with congenital heart disease and the children without congenital heart disease.Methods:We collected 97 cases of children's peripheral blood of congenital heart disease which included atrial septal defect(ASD),ventricular septal defect(VSD)and patent ductus arteriosus(PDA)as case group and 50 cases of children peripheral blood of non-congenital heart disease children as control group.Then,the total RNA was extracted from peripheral blood leukocyte.The mRNA expression of DNMT1 and DNMT3Bof the two groups were detected by real-time RT-PCR.Results:The DNMT mRNA expression of the white blood cell showed that the DNMT1 and DNMT3Bexpression of the case group were higher than the control group(P〈0.01).Conclusion:The expression rising of DNA methylation transferase may play an important role in the congenital heart disease.
