一种淫羊藿多糖对大鼠慢性疲劳的改善作用与代谢通路研究

摘　　要：从淫羊藿叶中提取其多糖的一个主要组分(herba epimedii polysaccharide,HEP),经理化性质鉴定HEP为一种蛋白结合多糖,其中总糖质量分数为89.5%,半乳糖醛酸质量分数为10.26%,蛋白质质量分数为4.39%。采用束缚、大强度运动、噪音与拥挤等多因素诱导建立一个大鼠的慢性疲劳综合征(chronic fatigue syndrome,CFS)模型,给服一定剂量的HEP,通过观察大鼠生长情况、饮食情况,并结合水迷宫实验、旷场实验和悬尾实验等行为学指标,评价HEP对CFS的治疗作用。最后采用代谢组学技术对大鼠尿液进行气相质谱法检测分析,寻找各组之间的差异代谢物及其代谢通路。结果表明,3组大鼠的尿液代谢物得到明显区分,安静组与CFS模型组对比,鉴定出11个潜在生物标志物;HEP组与CFS模型组对比,鉴定出5个潜在生物标志物。HEP主要通过调节CFS大鼠体内的HPA轴功能异常和精氨酸、脯氨酸代谢2条代谢途径治疗CFS。A main component HEP, extracted from the leaves of epimedium, is a protein bound polysaccharide which identified by its physical and chemical properties. The total sugar mass frac tion of HEP is 89. 5%, the content of galactose acid is 10. 26%, and the protein content is 4.39%. A chronic fatigue model was established by using multiple induced factors： the con straint, running platform, and congested and noisy environment. Giving certain amount of HEP, through the observation of rats＂ weight, intake of food and drinking water quantity, combin with general scoring scale and behavioral science experiment（water maze, open field, hanging lng tail test）, the HEI^s impact on CFS model was evaluated. Finally, the urinary of chronic fatigue rats was detected by GC-MS Technology of metabolism. And the difference of each group＇s metabolite and metabolic pathway were found out . The results showed that the urine metabolites of the 3 groups were well differentiated. A total of 11 metabolites were obtained between the CFS mod el and control groups with 5 potential metabolic biomarkers. And HEP mainly effects on chronic fatigue rats through two pathways： the disorder of HPA and the metabolism of arginine and proline.
