D-手性肌醇对IR-HepG2细胞葡萄糖消耗量的影响

摘  要:目的建立HepG2胰岛素抵抗细胞(IR-HepG2)模型,探讨D-手性肌醇(DCI)对IR-HepG2细胞葡萄糖消耗量的影响。方法采用细胞增殖与毒性检测(CCK-8)法,观察DCI、高糖对HepG2细胞活力的影响;通过高糖持续作用、胰岛素刺激诱导HepG2细胞建立IR-HepG2模型,葡萄糖氧化酶法鉴定模型是否制备成功并检测正常HepG2细胞葡萄糖消耗量的变化和DCI对IR-HepG2葡萄糖消耗量的影响。结果 0-0.5 mmol/L浓度的DCI对HepG2细胞活性没有影响,1.0-32.0mmol/L浓度的DCI对HepG2细胞活性产生抑制作用,差异有显著性(F=36.47,P〈0.01)。35-55 mmol/L的葡萄糖对HepG2细胞活性没有影响,而65 mmol/L的葡萄糖对HepG2细胞活性产生抑制作用,差异有显著性(F=18.88,P〈0.01)。DCI在不影响细胞活性浓度范围内可明显促进IR-HepG2的葡萄糖消耗量,差异有显著性(F=172.67,P〈0.01)。结论 DCI可明显增加IR-HepG2模型葡萄糖的消耗量,对IR-HepG2模型胰岛素抵抗有显著的改善作用。Objective To create a HepG2 insulin-resistant cell (IR-HepG2) model and explore the impact of D-chiro-inositol (DCI) on extracellular glucose consumption in insulin-resistant HepG2 (IR-HepG2) ceils. Methods Employing cell proliferation and toxicity test (CCK-8), the effects of DCI and high glucose on the viability of HepG2 cells was determined. An IR- HepG2 model was created through continuous action of high glucose and of insulin stimulation on HepG2 cells. Whether the model was successfully established was determined using glucose oxidase method, and the changes of glucose consumption in normal HepG2 cells and the influence of DCI on glucose consumption in IR-HepG2 cells were detected. Results The concentration of 0-0.5 mmol/L of DCI did not impact the viability of HepG2 cells, and that of 1.0-32 mmol/L produced inhibitory effect, the difference was significant (F = 36.47 ,P〈0.01). 35-55 mmol/L of glucose had no effect on the activity of HepG2 cells, and that of 65 mmol/L inhibitaed the activity of HepG2 cells, the difference was significant (F = 18.88, P〈0.01). Within the range of concentration, DCI could significantly promote the glucose consumption of RI-HepG2 cells (F=172.67,P〈0.01). Conelusion D-chiro-inositol can markedly increase the consumption of glucose in IR-HepG2 model, which improves insulin resistance in IR-HepG2 model.
