摘 要:目的从分子生物学水平揭示穴贴对牙髓的保护作用。方法选取60只Wistar大鼠,所有鼠牙牙体、牙列完整,无龋病,咬合关系正常。适应性喂养1周后,随机均分为空白组、模型组、阳溪组、颊车组。空白组:正常喂养,不实施任何干预手段;模型组:采用开髓封内毒素法(LPS)造急性牙髓炎模型;阳溪组:造急性牙髓炎模型,取远端同侧阳溪穴进行穴位贴敷;颊车组:造急性牙髓炎模型,取近段同侧颊车穴进行穴位贴敷,牙痛穴贴为自制。采用免疫组化法检测Notch1、Notch2蛋白在第1、3、5天的表达。结果 Notch1蛋白:穴贴治疗第1天,空白组表达量低于模型组、阳溪组、颊车组(P<0.05,P<0.01),第3天,模型组蛋白表达量低于阳溪组和夹车组,但高于空白组(P<0.05,P<0.01)。第5天,空白组与其它各组比较表达量降低(P<0.01)。Notch2信号蛋白:穴贴治疗第1天,穴贴各组与模型组和空白组两两比较表达增高(P<0.05,P<0.01)。第3天,模型组与其它各组比较,表达高于空白组,低于穴贴治疗各组(P<0.05,P<0.01)。第5天,空白组与其它各组比较表达量降低(P<0.01)。结论牙痛穴贴可通过缩短Notch信号蛋白表达上调时间,提高急性牙髓损伤后的自身修复能力,间接提高牙髓干细胞活性。Objective To explore the protective effect of dental pulp by toothache point stick and its molecular bio-logical mechanism. Methods Sixty Wlstar mouse were caries, normal occlusion. The rats were randomly divided into blank group, model group, Yang-xi group, Jia-ehe group, each group including 15 rats. The blank group : normal feeding and don' t implement any intervention. The model group : made the acute pulpitis rats with opening sealing and put in lipopoly saceharides. The Yang-xi group : made the acute pulpitis rats, and sticking aeupoint of the same side Yangxi point. The Jia-ehe group: made the a- cute pulpitis rats, and sticking acupoint of the same side Jiache point. The toothache point sticks were homemade. Immunohistoehemistry was used to detect Notehl protein and Notch2 protein expression in acute pulpitis rats at the first day, the third day and the fifth day. Results The Notehl protein, at the first day that treated with toothache point stick, the blank group was lower expression than model group, Yang-xi group and lia-ehe group (P 〈 0.05, P 〈0.01). The third day, the model group was lower expression than Yang-xi group and Jia-che group, but higher than blank group (P 〈 0. 05, P 〈 0.01 ) . The fifth day, the blank group was lower expression than the other groups (P 〈 0. 01 ). The Notch2 protein, at the first day that was treatment by toothache point stick, the Yang-xi group and Jia-che group were all higher expression than model group and blank group (P 〈0. 05, P 〈0.01 ) . The third day, the model group was higher expression than blank group, and the lower expression than Yang-xi group and Jia-che group (P 〈0.05, P 〈0. 01 ). The fifth day, the blank group was lower expression than the other groups ( P 〈 0.01 ). Conclusion The toothache point stick can indirect improve dental pulp stem cells activity, through accelerating the speed of Notch signal protein impressed time.
