摘 要:目的:采用HPLC法检测不同地区所产虎杖饮片中的4种有效成分(虎杖苷、白藜芦醇、大黄素和大黄素甲醚)的含量,为鉴别虎杖药材的真伪优劣提供依据。方法:色谱条件为色谱柱Agilent Zorbax SB-C18柱(4.6mm×250mm,5μm);流动相为0.1%甲酸水溶液(A相)-乙腈(B相),梯度洗脱(0~15min,20%B相;15~20min,20%~40%B相;20~30min,40%~70%B相;30~35min,70%~80%B相);检测波长306nm;流速1ml/min;柱温30℃;进样量为10μl。结果:虎杖饮片中的有效成分虎杖苷、白藜芦醇、大黄素和大黄素甲醚分离良好,精密度、重复性和稳定性均符合标准。虎杖苷在9.34~83.74μg/ml、白藜芦醇在4.07~36.65μg/ml、大黄素在3.74~33.62μg/ml、大黄素甲醚在3.62~32.54μg/ml范围内线性关系良好。虎杖苷、白藜芦醇、大黄素和大黄素甲醚的加样回收率分别为(98.21±1.086)%、(97.70±0.802)%、(97.58±1.005)%和(97.38±0.714%)(n=6)。结论:该方法快捷简便、稳定可靠,可用于虎杖饮片中有效成分的含量测定。Objective: To establish a HPLC method for content determination of four active ingredients(polydatin,resveratrol,emodin and physcione) in Polygonura cuspidaturn from different regions, so as to provide reference for quality identification of Polygonum cuspidatum. Methods: An Agilent Zorhax SB-C18 column(4.6 mm×250mm,5μm) was employed with the mobile phase consisting of 0.1% formic acid(phase A)-acetonitrile(phase B), gradient elution(0-15 min, 20% phase B; 15- 20 min,20%-40% phase B:20-30 min,40%-70% phase B:30-35 min,70%-80% phase B) at a flow rate of 1.0 ml/min. The detection wavelength was 306 nm, the column temperature was 30 ℃, and the sample size was 20 μl. Results: Polydatin, resveratrol, emodin and physcione displayed good linearity (r= 0. 999 9) within the ranges of 9. 34-83.74 μg/ml, 4.07-36.65 μg/ml, 3. 74- 33.62μg/ml and 3.62-32.54μg/ml, respectively. The recovery rates of polydatin, resveratrol, emodin and physcione were respectively (98. 21± 1. 086)%, (97.70 ±0. 802)%, (97. 58±1. 005)% and (97.38 ± 0. 714%)(n= 6), respectively. Conclusion: The method is rapid, simple, stable and reliable, and could be used for the determination of active constituents in Polygonurn cuspidaturn.
