GM1对脑出血大鼠LINGO-1表达的影响

摘  要:目的观察脑出血后LINGO-1表达的变化,探讨GMl对脑出血后LINGO-1表达的影响。方法将72只SD大鼠随机分为假手术纽、模型组和GM1治疗组,选取造模后1d、3d、7d和14d为观察点。制备脑出血模型,Longa评分法评价神经功能缺损程度,RT-PCR法检测LINGO-1mRNA的表达,Westernblot法检测LINGO-1蛋白的表达。结果模型组Longa评分7d最高,14d出现下降;LONG-1mRNA3d表达最高,7-14d出现下降;LONG-1蛋白7d到达高峰,14d出现下降。GM1治疗组在14d Longa评分较模型组下降(P〈0.05);在7d和14d LINGO-1mRNA表达较模型组下降(P〈0.05):在14d LINGO-1蛋白表达较模型组下降(P〈0.05)。结论脑出血后LINGO-1表达明显上调,呈先上升后下降的变化规律。GM1可以降低LINGO-1 mRNA和蛋白的表达及神经功能评分,这可能是GM1促进脑出血后神经功能恢复的机制之一。Objective To explore the effect of monosialotetrahexosyl ganglioside on expressions of LINGO-1 in rats with intracerebral hemorrhage ( ICH ) . Methods 72 SD rats were randomly divided into sham operation group, model group and monosialotetrahexosyl ganglioside group. Observation was made at 1, 3, 7, 14d after ICH. Neurological functional recovery was evaluated by Longa's score; RT-PCR and Western blot were applied to detect the changes in the expressions of LINGO-1 mRNA and protein. Results Longa's score and LINGO-1 protein were decreased at 14d after ICH in monosialotetrahexosyl ganglioside group compared with those in model group ( P〈0.05 ) . The LINGO-1 mRNA was decreased at 7d and 14d after ICH in monosialotetrahexosyl ganglioside group compared with model group ( P〈0.05 ) . Conclusions The decreased expressions of LINGO-1 and neurological score induced by monosialotetrahexosyl ganglioside may be one of the mechanisms of neural function recovery after ICH.
