摘 要:[目的]优化美洲大蠊蛋白质的提取方法,并研究美洲大蠊蛋白质对小鼠伤口愈合的影响。[方法]采用碱提法、盐提法和酶提法对美洲大蠊蛋白质进行分离和提取,测定不同方法提取美洲大蠊蛋白质的含量,优化美洲大蠊蛋白质的提取工艺;并用提取的美洲大蠊蛋白质进行小鼠伤口愈合试验,研究美洲大蠊蛋白质对小鼠伤口愈合的影响。[结果]在碱浓度为1.5%、提取温度为80℃、提取时间为1.0 h时,美洲大蠊蛋白质提取率最高;在盐浓度为1.0%、提取温度为40℃、提取时间为2.0 h时,美洲大蠊蛋白质提取率最高;在酶占样本比为0.6%、酶解温度为45℃、酶解时间为2.0 h时,美洲大蠊蛋白质提取率最高。小鼠伤口愈合试验表明,美洲大蠊提取的蛋白质可以促进伤口愈合,且愈合时间比正常情况下缩短近一半。[结论]碱液浓度、盐浓度和酶浓度对美洲大蠊蛋白质提取的影响最大。3种方法提取美洲大蠊蛋白质的效果从大到小依次为酶提法、碱提法、盐提法,即美洲大蠊蛋白质提取的最优方法为胰蛋白酶提取法。美洲大蠊蛋白质对小鼠伤口愈合有促进作用。[ Objective ] To optimize extraction method of Periplaneta americana protein and study its effect on wound healing of mice. [ Method ] The protein was seperated and extracted from P. americana by alkali extraction, salt extraction and enzyme extraction, protein content ex- tracted by different methods were determined, the extraction techniques were optimized ; effects of protein from P. americana on wound healing of mice were studied. [ Result] The protein yield was highest under three conditions: alkali concentration 1.5% , extraction temperature 80 ℃, extraction time 1.0 h; salt concentration 1.0% , extraction temperature 40 ℃, extraction time 2.0 h; proportion of enzyme 0.6% , enzymolysis temperature 45 ℃ , enzymolysis time 2.0 h. Protein from P. americana could promote mice wound healing, and healing time was shortened by nearly half. [ Conclusion] Alkali liquor concentration, salt concentration and enzyme concentration have greatest impact on pro- tein extraction. The order of extraction effect is enzyme extraction, alkali extraction, salt extraction, namely the optimal method for extracting protein from P. americana is trypsin extraction. Protein from P. americana can promote mice wound healing.
