摘  要:目的:研究香砂六君丸对肥胖大鼠胃黏膜JAK2和STAT3表达的影响,探讨该方控制肥胖人群罹患胃癌风险的可能性。方法:40只雄性SD大鼠,随机分为对照组、肥胖组(模型组)、香砂六君丸低剂量治疗组(香低组)、香砂六君丸中剂量治疗组(香中组)和香砂六君丸高剂量组治疗组(香高组),每组8只;全自动生化分析仪检测血脂;ELISA法检测血清超氧化物歧化酶(SOD),丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-Px)活性;免疫组化法检测胃黏膜磷酸化(p)-JAK2/STAT3表达。结果:与对照组比较,模型组的总甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)和MDA水平以及p-JAK2、p-STAT3表达均显著增加,但SOD和高密度脂蛋白(HDL)水平以及GSH-px活性均明显下降。与模型组相比,香低、中和高组的TG和LDL水平显著下降,而SOD水平和GSH-px活性明显上升;香中组和香高组MDA水平和p-JAK2、p-STAT3表达均显著下降;香高组HDL水平明显上升。结论:香砂六君丸能够调控血脂,对抗氧化应激,抑制胃黏膜JAK2/STAT3通路,可能具有降低肥胖人群罹患胃癌的作用。Objective: To study the effect of Xiangsha Liujun Pills ( XLP ) on expressions of JAK2 and STAT3 in gastric mueosa of obesity rats, exploring the possibility of XLP controlling the risk of obesity people developing gastric cancer.Method: A total of 40 male SD rats were randomly divided into the control group, obesity group ( model group ), XLP low-dose treating group ( XLG ), XLP middle-dose treating group ( XMG ) and XLP high-dose treating group ( XHG ), 8 rats in each group.Antuomatic biochemical analyzer was used to check serum lipid profile.ELISA was chosen to check serum levels or activity of SOD, MDA and GSH-Px.Immunohistochemistry was employed to detect expressions of p-JAK2 and STAT3 in gastric mueosa.Resnlt: Compared with those of the control group, levels of TG, TC, LDL and MDA and expressions of p-JAK2 and STAT3 were all increased, but SOD and HDL levels and GSH-px activity were significantly decreased in the model group.Compared with those of the model group, TG and LDL levels were decreased, and SOD level and GSH-px activity were markedly increased in XLG, XMG and XHG.MDA level and p-JAK2/STAT3 expressions were decreased significantly in XMG and XHG.HDL level was improved obviously in XHG.Conclusion: XLP can regulate serum lipid profile, opposite oxidative stress and inhibit JAK2/STAT3 pathway of gastric mucosa in obesity rats, suggesting it might be used to control the risk of obesity people developing gastric cancer.
